Research purpose:This aims are to observe the begins with prehypertension swimming movement influence the expression of CTRP9 and influence on vasodilatation function and specific molecular mechanism,explore the signaling pathways in which the role of.Methods:using 20 male of WKY rats and 24 male of SHR rats,which is 4 weeks,later adaptability to feed them one week,the WKY were randomly divided into two groups,namely the quiet control group(W,n = 9),the aerobic exercise group(WE,n =11),SHR were randomly divided into two groups,namely the quiet control group(S,n=10),the aerobic exercise group(SE,n = 14).The exercise group had 10 weeks of swimming training,five days a week,and the first week of training,10min,than 20min the next day,30min on the third day,45min on the fourth day and 60min on the fifth day.Starting from second week,the swimming activity of 60min was performed every day for five days and lasted nine weeks.Ten weeks after swimming movement,using the determination of the carotid artery intubation in rats blood pressure and heart rate,the determination of serum level of CTRP9 by ELISA competition law,and with NO detection kit will measure the levels of serum NO,microvascular ring determination of secondary and tertiary mesenteric artery diastolic properties of mesenteric artery CTRP9 protein immune blot method,AdiopRl,AMPK,the expression of eNOS protein,paraffin section observation group artery morphological differences.Research results:(1)After 10 weeks of swimming,the systolic and diastolic blood pressure in the SE group decreased significantly compared with group S,and the weight and average arterial pressure in SE group were no different from that of group S.The weight of our group was significantly lower than that of W group,and the diastolic pressure,systolic blood pressure and average arterial pressure in the group were no different from that of group W.The systolic blood pressure and average arterial pressure in group S were significantly higher than that in the W group.The group weight of group S was significantly lower than that in the W group,and the diastolic blood pressure and the heart rate of group S were no different from that in the W group.It is suggested that long-term swimming can reduce the blood pressure of rats with high blood pressure,which can prevent the normal weight gain of normal rats.Long-term swimming exercise can reduce the weight of rats.(2)After 10 weeks of swimming,the concentration of serum CTRP9 in SE group was significantly higher than that in group S.The concentration of serum CTRP9 in the group was significantly higher than that in group W.The concentration of serum CTRP9 in S group was lower than that of W group.It is suggested that the concentration of serum CTRP9 decreased in hypertensive state,and the long-term aerobic exercise may improve the concentration of serum CTRP9 in hypertensive rats.After 10 weeks of swimming,the serum NO concentration in SE group was significantly higher than that in group S.There was NO significant difference in serum NO concentration compared with W group.The serum NO concentration in S group was significantly lower than that in W group.The serum NO levels in rats were significantly reduced in hypertensive rats,and long-term aerobic exercise could improve serum NO level in rats with high blood pressure,but there was NO significant difference in serum NO content in normal rats.(3)The mesenteric vascular ring in PE(10-5 M)preshrunk,with a concentration dependent diastolic response to the SNP(10-9-10-5 M),the SE group,group S,WE group,and W group diastolic ability were no different.Mesenteric vascular ring in PE(10-9 to 10-5 M)after pre shrinking,ACH(10-9-10-5 M)can induce concentration dependence in diastole,SE group of arteriole endothelium-dependent relaxing function compared with S group increased significantly;There was no significant difference between the endothelial dependent diastolic function and the W group.The endothelial dependent diastolic function of the microartery of S group decreased significantly compared with W group.Add L-NAME(10-4 M)to pre-incubate blood vessels for 20min,and use PE(10"5 M)to precollect micro vascular ring,then add ACH(10-9-10-5 M)to induce vasodilation.Compared with the control group,the early incubation of 1-name was decreased in all groups of mesenteric artery diastolic reactions,and the decrease of group S was significantly lower than that of other groups.It is suggested that the diastolic function of the microarterial endothelium in rats with hypertensive condition decreased,and the long-term swimming exercise could improve the diastolic function of the microarterial endothelium in rats with hypertensive rats.(4)Ten weeks after swimming,SE group rat arterial tissue CTRP9,AdipoR 1,AMPK and eNOS protein expression significantly increased compared with S group,WE group rat arterial tissues CTRP9,AdipoR 1,AMPK and eNOS protein expression significantly increased compared with W group,the group S rat arterial tissue CTRP9,AdipoR 1,AMPK and eNOS protein expression compared with W group dropped significantly.It is suggested that hypertension causes the expression of the expression of CTRP9,AdipoR 1,AMPK and eNOS proteins.Long-term swimming exercise can effectively increase the expression of CTRP9,AdipoR 1,AMPK and eNOS.(5)HE staining could observe that compared with W group,the smooth muscle cells in the group WE group were smaller in size,with a spindle shape and a long,rod-shaped nucleus.Compared with group S,the rats in the SE group were thinner and the smooth muscle cells were smaller and smaller.Compared with W group,the wall of rats in group S was thickened,and the middle level smooth muscle cells were hypertrophy and nuclear.It is suggested that the blood vessel wall thickens and the smooth muscle hypercell hypertrophy,and the swimming exercise can improve the vascular disease in the condition of hypertension.Research conclusions:the basis of aerobic exercise can effectively reduce SHR rats blood pressure,strengthen the function of mesenteric arterial vasodilation,CTRP9 is probably one of the important mechanism,which may be through blood vessels AdipoR 1/AMPK/eNOS/NO way to work.
