OBJECTIVE:At present, the hypertensive diseaseas, as one of cardiovascular diseases system, endangers the health and life of human being seriously. The hypertension diseases associate with insulin resistance, vascular endothelial cells, and metabolic disorders as well. The treatment of hypertension in the scientific field is now an important task. Exercise can improve the function of cardiovascular system in spontaneously hypertensive rats, from which the skeletal muscle grows and also the quality of the bone improves. Rizzoni spent eight weeks to twelve weeks old of hypertensive rats by taking antihypertensive drugs, used the Western Blot method for detecting, and found the improved scarce spontaneously hypertensive keletal muscle microvascular. Previous studies found that aerobic exercise can increase the microvascular density of the gastrocnemius muscle of spontaneous hypertensive rat, however, the research of reduced situation of flounder vessels in spontaneous hypertensive rats and whether exercise is good for the improvement is still lacked. This paper gives discussion about the effects of exercise in spontaneously hypertensive rat soleus blood vessels and its related mechanism, which will be helper for preventing the hypertension disease and giving advice to experimental and theoretical basis.Research MethodsFour weeks old male rat used as the research object in this research, the number of rat is 25 and divids into three groups. Normal pressure rats used as contrast group and the number was nine (Wistar kyoko rat Wistar kyoko, WKY). Spontaneously hypertensive rats set as a group and the number was eight (Spontaneously hypertensive rat, SHR), spontaneous hypertension rats were stimulated by swimming and the number is eight (Spontaneously hypertensive rat+Exercise, SE). The stimulation of rats was drived by swimming. The experimental was performed after one week’s adaptive training. The experitment was carried at the temperature of 32-34 degrees Celsiu, the ennveriment was required to be quiet and peace. In the first and second week, the rats were exercised for sixty minutes per week, while from week 3 to week 6, the exercise was performed ninety minutes, six every day. In week 7 and week 8, the exercise increased to a hundred and twenty minutes, every six day. The specimens were made immediately after the training. HE staining was observed in the soleus form, while the immunohistochemical detection was detectived by the expression of CD31, Wester Blot methods was to test VEGF protein, PI3K, eNOS, Akt protein expressions.Experimental results:1 HE staining results showed that compared with normal blood pressure rat group, the fiber cross-section area of spontaneous hypertension in rat in skeletal muscle was reduced compared with soleus muscle and had significant difference (P< 0.01); The cross-sectional area of muscle fiber in the exercise group of spontaneously hypertensive rats increased significantly (P< 0.01) compared with hypertension rats. This indicated that exercise can increase the muscle cross-sectional area of muscle fiber in SHR.2 The number of microvasculare signed by CD31 showed that compared with WKY, soleus in SHR decreased; whereas compared with SHR, the number of microvasculare solesus signed by CD31 in SE increased(P<0.01). This indicated that aerobic exercise improved sparse of the blood vessels of spontaneously hypertensive in rats.3 The expression of vascular endothelial growth factor (VEGF) protein showed that VEGF in the group of spontaneous hypertension was less than that in normal pressure(P< 0.01), while this raised after exercise in hypertension rat compared with spontaneously hypertensive rats (P< 0.01). That experiment implied that exercise can increase the expression of VEGF in spontaneously hypertensive rat soleus.4 The protein expression of PI3K/Akt/eNOS in spontaneous hypertension group was cut down than the normal blood pressure group in rat skeletal muscle (P< 0.01), but exercise in hypertension increased than hypertension group (P< 0.05). This proofed that exercise can increase the expression of Pl3K/Akt/eNOS in spontaneously hypertensive rat soleus. immediately after the training.
