Obstructive sleep apnea syndrome(OSAHS)is caused by repeated chronic intermittent hypoxia(CIH),)during sleep,which results in multiple organ damage and great damage to personal health and socio-economy.It has been found that exercise has significant protective effects on multiple organ function damage,especially kidney damage in OSAHS patients,but the mechanism is unknown.Object:To investigate the effects of aerobic exercise on renal oxidative stress and its regulatory pathway in intermittent hypoxia mice.Methods:40 6-week-old male C57B1/6 mice weighing 20g/25g were selected.The mice were randomly divided into 4 groups:control group(C group),exercise control group(E),intermittent hypoxia group,(H),intermittent hypoxia exercise group),(HE),group(10 mice in each group).The chronic intermittent hypoxia device controlled by a computer was used to model CIH.Group C was fed for 5 weeks,group E was subjected to adaptive treadmill exercise for one week under normoxic condition,and 60 min treadmill exercise was performed daily from the second week.Group H received intermittent hypoxia treatment for 8 hours per day for 5 weeks.HE group received treadmill adaptive exercise after CIH treatment every day in week 1,starting from week 2,group H received intermittent hypoxia treatment for 8 hours per day for 5 weeks.After 8 hours of CIH treatment every day,treadmill exercise was performed for 60 minutes,adaptability and formal exercise for 5 weeks.At the end of the experiment,the morphological changes of renal tissue were observed by hematoxylin-eosin staining staining.The glomerular filtration function was measured by Elisa.The(MDA)content of malondialdehyde(MDA)in serum and renal tissue was measured by Elisa.The level of superoxide dismutase(SOD)was measured by hydroxylamine method,and the protein expression of p-Nrf2 and HO1 in the signal pathway was detected by Western Blot.Results:(1)The results of hematoxylin-eosin staining showed that CIH caused necrosis or swelling of glomeruli and widened or narrowed the space of cystic cavity in mice.Aerobic exercise significantly improved the pathological changes of renal tissue in HE group.(2)Compared with group C,the level of plasma CYS-c in group H was significantly higher than that in group H(P<0 01),and the level of CYS-c in group HE was significantly lower than that in group H(P<0 01).Similarly,the plasma ? 2-MG level in group H was significantly higher than that in group C(P<0.05),and the plasma ? 2-MG level in group HE was significantly lower than that in group H(P<0.05).(3)The content of SOD in plasma and SOD in renal tissue in group H was significantly lower than that in group C(P<0 05),and that in group H was significantly lower than that in group C(P<0 05).The content of SOD in plasma and the content of SOD in renal tissue in HE group were significantly higher than those in H group(P<0.01).Similarly,the levels of MDA in plasma and MDA in renal tissue in group H were significantly higher than those in group C after chronic intermittent hypoxia(P<0 05),and that in group H was significantly higher than that in group C(P<0 01).The plasma MDA content and renal tissue MDA content in HE group were significantly lower than those in H group(P<0.05).(4)Compared with C group,the phosphorylated protein expression level of Nrf2 in H group was significantly lower than that in C group(p<0.01).After aerobic exercise,the expression level of p-Nrf2 in HE group was significantly higher than that in H group(p<0.01),and the expression level of HO1 protein in).H group was significantly lower than that in C group(p<0.01).The expression level of HO1 protein in HE group was significantly higher than that in E group after aerobic exercise(p<0.01).Conclusion:(1)Aerobic exercise can improve renal oxidative stress in intermittent hypoxia mice.(2)Aerobic exercise may play a protective role in renal function by regulating the level of antioxidant stress in CIH mice activated by Nrf2,HO1 and other signaling molecules.
