ObjectiveThe thesis aims to, in terms of anti-carbonyl stress, conduct a primary research about anti-carbonyl stress defense system by employing amino-containing biological small molecules(taurine、leucine、isoleucine、 valine).It is also expected to probe into the protective effect that amino acid mixture has on after aerobic exercise rats by refering to related research results. From the perspective of free radical metabolism,the research was designed to provide rats with combine to supply and discern to supply extrinsic source taurine and the series of amino acid mixture. By making use of model of rat aerobic exercise.Materials and methodsThe experimental subjects,40SD male rats, were randomly divided into group control(C),exercise group(S), exercise with taurine(ST), exercise with the series of amino acid (SB), exercise with taurine and the series of amino acid mixture(STB) with8in every group. Group C and S were injected Nacl with dosage of0.1ml/10g per day, while group ST,SB and STB were injected taurine,the series of amino acid and amino acid mixture,respectively with the same dosage. All the exercising group experienced aerobic exercise for8weeks according to the exercise program. Blood, skeletal muscle and brain tissue were taken out after the program. Then we tested the weight of rats,LA content,skeletal muscle and brain tissue of rats,SOD,CAT,GSH-Px,MDA content and protein carbonyl, and observed the ultrastructure of skeletal muscle and brain tissue.Research resultsWeight and test results of blood lactateThe weight of rats in all groups gained significantly after8week aerobic exercise. There was great weight difference between before the experiment and after wards. Compared group c,the average weight of group ST,SB and STB dropped obviously (P<0.05). There was no distinct difference in weight between group ST and group C,and between exercise with amino acid and exercise group. In comparison to group SB and STB,the blood lactate in rats of group S decreased clearly c(P<0.05and P<.01).The effect of amino acid mixture on aerobic exercise rat skeletal muscle tissueSOD,GSH-Px, and CAT activity of skeletal muscle tissue of group ST,SB and STB distinctly exceed that of group S and C (P<0.05and P<0.01),while group STB overtopped group SB and ST (P<0.05and P<0.01) in all these activities. There was a tendency of SOD activity of group S exceeding group C. With respect to skeletal muscle tissue MDA content and protein carbonyl,group ST,SB and STB were under group S and C group(P<0.05and P<0.01), S was below group C (with no statistic significance).however, the group S of protein carbonyl was lower than that of group C(P<0.05).MDA content of group STB was remarkbly lower than group SB (P<0.05).The effect of amino acid mixture on aerobic exercise rat brain tissueSOD,GSH-Px, and CAT activity of brain tissue of group ST,SB and STB distinctly exceed that of group S and C (P<0.05and P<0.01), SOD,GSH-Px activity of group S distinctly exceed that of group C(P<0.05). while group STB overtopped group SB and ST in all these activities. With respect to brain tissue MDA content and protein carbonyl,group ST,SB and STB were under group S and C group(P<0.05and P<0.01).however, the group S of MDA content was lower than that of group C(P<0.05).MDA content of group STB was remarkbly lower than group SB and ST(P<0.01) Results from observation in HE staining of skeletal muscle of ratsBy light microscopy,C rats skeletal muscle crosscut slice of visible muscle cells are in the form of polygons nest distribution, muscle nuclear distributed around muscle plasma (under the muscle membrane), normal size, no swelling, no solid shrink.,and we can’t see obvious blood vessel change and obvious inflammatory reaction in the muscle interstitial; by observing the crosscut slice of S rats,we can see that the muscle fiber increased, and we can’t see the muscle dissolved and muscle swelling, as well as the muscle wire fracture; the skeletal muscle form of SB group, and ST group and the STB rats didn’t show abnormal.Results from observation in HE staining of brain tissue of ratsBrain tissue structure of the rats in group c was clear and complete through polarizing grass. Its nerve cells were intensive, orderly arranged and gap-dense without any edema. The gaps surrounding the cell of rats in group s were normal and regularly formed. It can be concluded that a few nerve cells disappeared and there was no edema. Group ST,SB and STB share similarities with group C.Conclusion(1) Taurine and a series of amino acid can reduce the accumulation of lactic acid in bodies. Amino acid mixture works better than the exclusive use of taurine and a series of amino acid;(2)8-weeks aerobic exercise helps to defence against carbonyl stress;(3)8-weeks aerobic exercise, combined with taurine,a series of amino acid and amino acid mixture, can enhance SOD,CAT and GSH-Px activity, strengthen antioxidant and ability of organism, and reduce the concent of MDA and protein carbonyl as well. It demonstrates than taurine,a series of amino acid and amino acid mixture can resist oxidation and anti-carbonyl stress, clear away reactive carbonyl species and oxidation stress, thus preventing from damage of carbonyl stress;(4)8-weeks aerobic exercise combined with amino acid mixture functions better than exclusive use of taurine and the series of amino acid.
