Abstract/Summary:
Avian influenza (AI) caused by avian influenza viruses H5N1 has been concerned as a threat to public health with pandemic potential. There is an urgent need to develop sensitive, specific and rapid detection methods for early diagnosis to lower the chances of AI spread and reduce the risk of AI development into an epidemic.;The viruses ranging from 100 to 107 EID 50/ml with a detection time of 1 h were incubated with the impedance measurement followed. The results indicated a minimum detectable avian influenza virus concentration of 103 EID50/ml. The biosensor was tested to be specific and sensitive.;Focusing on optimizing the detection system for further application, the main parameters were systematically investigated. The signal amplification, the medium for impedance measurement and the detection time which was defined as the incubation time of AIV sample were studied.;Overall, the immuno-detection of avian influenza virus H5N1 based on the impedance measurement using interdigitated array (IDA) microelectrodes was effective method with high sensitivity and rapidity. The optimization of the sensing system improved its performance and enabled its further applications.;In this research, an impedance biosensor was demonstrated and evaluated for the detection of avian influenza virus (AIV) H5N1 based on the interdigitated array (IDA) microelectrode, using the immunosensing technique. At the same time, for improving the performance of the biosensor, the mainly factors affecting the performance of the biosensor were investigated and the parameters were optimized to achieve high sensitivity with short detection time.
