Background and objectiveMusic therapy discipline has been booming since it founded in the 1940s,established and developed many practice and theory genres to explore the mechanism of music therapy from different angles,such as psychological physiology and behavior,and accumulated a lot of research results.On the one hand,to prove the effectiveness of music therapy,on the other hand explains the principle of the music therapy from different angles.In addition,music therapy in clinical also has a good performance,music can improve stroke patients with movement and cognitive memory,sensory processing and visual attention ability;music has blood pressure lowering effect,and the musical effect of the different frequency has obvious difference;the music can help patients with Alzheimer’s disease(AD)enhance memory.Music also can help the patients to relieve pain and reduce anxiety during the process of surgery and the treatment;the music can improve gait and cognitive states of Parkinson’s disease(PD)patients;music therapy is widely used in the hospice care.And in the past,most of the research is based on whole animals,with the complete auditory system as a precondition to explore and think about the principle of music therapy,which in a certain extent is because many researchers tend to define music therapy as a category of psychological treatment and consider that the therapeutic effect is based on the music by causing mental and emotional fluctuations and then change the physiological state.We do not deny the effects of psychological intervention effect from music,it is worth pondering that whether music therapy must be based on the psychological effect,whether music can directly induce physiological changes,what is the mechanism,to solve these puzzles will be conducive to better guide and the development of music therapy.This study is to explore the possible mechanism of music therapy on the cellular level through researching the intervention effects of music on non-auditory cells in vitro.1.We try to study the direct effect of music in different non-auditory cells,and to practice music therapy research into cellular and molecular levels,in order to provide more accurate and reliable theoretical basis and scientific guidance to music therapy prescription.2.In addition to consider music therapy prescription,and we need to think about dose and the question of whether or not to have side effects.3.By studying the effects and value of sonic physical stimulation during music therapy in order to find more economical and safe treatment.MethodCultured 24h newborn SD rats’ primary cortical astrocytes in vitro;identify astrocytes by using immunofluorescence staining method;cultivation of Neuro-2a and HUVEC cell line;Isolation and extraction of lymphocytes;observe the location distribution and size of cells cloned island by inverted microscope;use the MTT assay to detect cell survival activity and astrocytes.Astrocytes’ growth tendency;observe the astrocytes’ultrastructural differences by transmission electron microscope;measure PaD2 and GFAP protein expression changes by Western blotting,and detect that whether after music intervention astrocytes can express neural stem cell marker nestin.Results1.The astrocytes of cerebral cortex cell from 24-hour rat are Identified as GFAP positive cells.2.By the inverted microscope we can find that music promotes the astrocyte cell proliferation,and promote lymphocyte clone island ready-made.3.Determined by MTT method to detect different cell survival activity increased significantly after music intervention,and this effect will change as time migration.4.To observe the in vitro growth curve of cells found that music can effectively promote the growth of astrocytes,and improves the cells to glutamic acid and glycine excited drug tolerance.5.Transmission electron microscopy to observe the ultrastructure of astrocyte found that music groups have more mitochondrial and synaptic populations.6.Western blot experiments found that astrocytes can express neural stem cell markernestin,and the amount of the expression of nestin will change by outside intervention,besides after music intervention PAD2 and GFAP protein expression will down-regulated in astrocytes.Conclusion1.It doesn’t need a complete system to perception and reaction to the music,different in vitro cultivation of cells can respond to music.2.Music can promote cell proliferation and growth.3.Music can improve the tolerance of cells excited poison.
