Objective:Glioma is the most common primary brain tumor,accounting for about 81% of malignant brain tumors.Current research on glioma lacks molecular markers that can accurately predict patient prognostic outcomes.Vacuole membrane protein 1(VMP1)has been regarded as an autophagy-related protein in previous studies,while recent studies have found that it is upregulated in breast cancer,acute myeloid leukemia,and pancreatic cancer.However,the role of VMP1 in glioma and its relationship with glioma have not been reported.Therefore,this study aims to investigate the relationship between VMP1 and glioma through bioinformatics and to infer how VMP1 affects the development of glioma.Methods:In this study,the sample data downloaded from The Cancer Genome Atlas(TCGA)was used as the study data set,and the sample data downloaded from Chinese Glioma Genome Atlas(CGGA)and Gene Expression Omnibus(GEO)were used as the validation data set.The data from the Chinese Glioma Genome Atlas(CGGA)and Gene Expression Omnibus(GEO)were downloaded as validation data sets,and the data were cleaned and organized to investigate the relationship between VMP1 and glioma from different perspectives,including different classification methods,survival analysis,and pathway enrichment,using statistical methods.Results:The results obtained by using bioinformatics techniques showed that the expression of the VMP1 gene is higher in gliomas than in normal brain tissue and increases with the malignancy of the pathological type,such as higher histological grading and higher WHO grading,and there may also be a relationship between VMP1 and key biomarkers in glioma research in recent years,such as the expression of VMP1 and Isocitrate dehydrogenase wild type(IDH-wt),O6-methylguanine-DNA methyltransferase(MGMT),and telomerase The results of the study also indicated that the expression of telomerase reverse transcriptase(TERT)was positively correlated with VMP.The results also point to the potential of VMP1 as a new biomarker to differentiate glioma from normal tissue or low grade glioma(LGG)from glioblastoma(GBM),as well as to be used independently to predict overall survival(OS),disease remission,and the number of gliomas.survival(OS),progression-free interval(PFI)and disease-specific survival(DSS).In addition,gene set enrichment analysis(GSEA)of VMP1 showed that samples with high VMP1 expression were significantly enriched in immune-related pathways,which further elucidates the possible biological processes involved in VMP1 in glioma.Conclusions:1.VMP1 is highly expressed in gliomas and increases with increasing malignancy of the glioma.2.VMP1 expression increases with histological grade and WHO grade.3.VMP1 is consistent with the trend of molecular biomarkers representing malignant.4.The potential pathways of VMP1 may be immune and proteasomerelated pathways.5.VMP1 expression is an independent risk factor for survival prognostic indicators OS,DSS and PFI,with higher expression representing a worse prognosis.6.VMP1 expression may have a strong diagnostic and predictive prognostic value for glioma and may be a new molecular biomarker for glioma.
