Objective:To bulid a regulatory network between lipid metabolism and susceptible gene in premature coronary heart disease(PCHD),and screen the meaningful early-warning and intervention targets.Find out the enrichment signal pathway related to the incidence of PCHD,explore the possible molecular biological process of the occurrence and development of premature coronary heart disease.Methods:(1)Blood is collected from patients with PCHD and healthy people,and serum samples are extracted from the two groups.(2)DNBseq platform is used to analyze the gene expression in serum samples from two groups.After the standardization of gene data,the reference genome was compared to screen out the differentially expressed genes between the two groups.(3)The differentially expressed genes were analyzed by Go function and KEGG pathway enrichment,then find the biological processes and functions involved in PCHD.Results:(1)A total of 33 different expressed genes were detected in the two groups of samples,including 17 up-regulated genes and 16 downregulated genes.(2)Among the 17 different up-regulated genes,miR-34a-5p,miR-1228-5p,miR-192-5p and miR-185-5p were the most significant.(3)GO functional cellular component analysis shows that the different expression genes were significantly enriched in cytoplasm,nucleoplasm,cytosol,cell projection,Golgi apparatus,cytoskeleton,nucleus,endoplasmic reticulum,cell junction,endosome;Molecular function analysis shows that the different expression genes were significantly enriched in protein binding,nucleotide binding,transferase activity,kinase activity,ATP binding,hydrolase activity,metal ion binding,RNA polymerase II proximal promoter sequence-specific DNA binding,DNA-binding transcription factor activity(RNA polymerase IIspecific),protein kinase activity;Biological process function analysis shows that the different expression genes were significantly enriched in positive regulation of transcription by RNA polymerase II,negative regulation of transcription by RNA polymerase II,phosphorylation,protein transport,multicellular organism development,intracellular signal transduction,Regulation of transcription by RNA polymerase II,cell cycle,nervous system development,intracellular protein transport.(4)KEGG pathway enriched analysis shows that different expression gene were involved in Metabolic pathways,MAPK signaling pathway,Ras signaling pathway,Adrenergic signaling in cardiomyocytes,Regulation of actin cytoskeleton,Axon guidance,Autophagy – animal,Platelet activation,Neurotrophin signaling pathway,Rap1 signaling pathway and play their biological functions.Conclusion:miR-34a-5p,miR-1228-5p,miR-185-5p,miR-192-5p may be related with PCHD?...
