Purpose: Pleckstrin homology domain containing O2(PLEKHO2)is a coding protein that promotes macrophage survival.However,the prognostic value of PLEKHO2 for gastric cancers and immune infiltration remains unknown.Methods: The Oncomine database analyze PLEKHO2 expression in diverse cancer types.Western blotting was used to verify the expression of PLEKHO2 in gastric cancer cells and tissues.We analyzed the relationship between PLEKHO2 expression and clinical survival data of gastric cancers via Kaplan-Meier plotter and UALCAN database.We evaluated the correlation between PLEKHO2 expression and immune infiltration of various cancers,using Tumor Immune Estimation Resource(TIMER)? GEPIA and tumor-immune system interactions database(TISIDB).Results: We found that PLEKHO2 was highly expressed in stomach adenocarcinoma(STAD).And high expression of PLEKHO2 was significantly correlated with poorer prognosis in patients with STAD via bioinformatics database.Furthermore.And high PLEKHO2 expression was obviously associated with tumor-infiltrating immune cell,especially of immune checkpoint molecules,including PD-1(programmed death-1),CTLA-4(cytotoxic T lymphocyte-associated antigen-4),LAG3(Lymphocyte-activation-gene-3)and TIM3(T cell immunoglobulin domain and mucin domain-3)in stomach adenocarcinoma.Conclusions: Our results suggest that PLEKHO2 expression can provide valuable information for the expression,prognosis,and influence of tumor immune infiltration in gastric cancer.In sum,PLEKHO2 can be used as an useful biomarker for the prognosis of patients with gastric cancer,and evaluate the immune cell infiltration of PLEKHO2 and predict the efficacy of immunotherapy in patients with gastric cancer.
