Objective: Using non-targeted metabolomics techniques,we investigated the differential metabolite changes and metabolic pathways in the healthy population before and after the short-term very low calorie restriction combined with exercise intervention,and analyzed and discussed some of the metabolic mechanisms underlying the effects of short-term very low calorie restriction combined with exercise intervention on the healthy population.Methods: A total of 36 subjects participated in a 5-day short-term very low calorie restriction combined with exercise intervention,in which short-term very low calorie restriction was defined as calorie intake <100 kcal per day,and low to moderate intensity aerobic exercise was performed during the calorie restriction period,including tai chi,fitness qigong,staking,and group activities.Body weight,heart rate and blood pressure were measured,small and dense low-density lipoprotein and fasting glucose were detected,and body mass index was calculated,and the differences of the above indexes before and after short-term very low calorie restriction combined with exercise intervention were analyzed using SPSS software.Plasma was collected from the subjects,and the differences in metabolites before and after the short-term very low calorie restriction combined with exercise intervention were analyzed using non-targeted metabolomics techniques.Results:(1)After the 5-day short-term very low calorie restriction combined with exercise intervention,subjects showed significant(P<0.05)reductions in body weight,body mass index and blood glucose,but no significant(P>0.05)changes in systolic and diastolic blood pressure.(2)A total of 38 differential metabolites were screened and identified in men and 33 differential metabolites were identified and screened in women before and after the short-term very low caloric restriction combined with exercise intervention in both positive and negative ion modes,and the main differential compounds produced were arachidonic acid,prostaglandins,caffeine,leucine,L-isoleucine,etc.The main secondary classifications of differential metabolites included lipids and lipid-like molecules,organic acids and their derivatives,and organic heterocyclic compounds.The main secondary classifications of differential metabolites included lipids and lipid-like molecules,organic acids and their derivatives,organic heterocyclic compounds,etc.(3)The results of significant enrichment pathways of differential metabolites(P<0.05)showed that there were three significant enrichment pathways in men and two in women before and after short-term very low caloric restriction combined with exercise intervention.Conclusion:(1)Short-term very low calorie restriction combined with moderate exercise intervention can significantly reduce the subjects' weight,BMI and blood glucose levels as well as regulate cardiovascular risk factors;(2)The main pathways involved include arachidonic acid metabolism,caffeine metabolism,valine,leucine and isoleucine biosynthesis,and short-term very low calorie restriction combined with exercise intervention can affect the metabolic status of healthy organisms to a certain extent.
