The polysaccharides of Sweet corncobs were extracted,separated and purified wirh m odern analysis methods.The experimental contents and results are as follows.The polysaccharides of Sweet corncobs were extracted by ultrasonic assisted with wat er as solvent.The optimum conditions were obtained through single factor experiment a nd response surface analysis The results showed that the optimum granularity,liquid rat io,ultrasonic time,ultrasonic temperature,power and extraction number,was 80 mesh,15:1(mL:g)45 min,25?,500 W,28 Hz and 1,respectively.The yield of polysacch arides up to 22.68%?Polysaccharide of sweet corncobs was decolored with the method of acticarbon.Using a single factor experiment and response surface method analysis was based on quadratic regression model,single factor experiment were carried out to determine amount of acticarbon,decolorization time and pH.The results indicated that the additive amount of acticarbon,adsorption time and pH was 2%,2.1h and 4,respectively.This method was compared with the method of hydrogen peroxide.The deproteinization with savage,there is no protein and nucleic acid determined through UV.Three components of Sweet corncobs polysaccharide were obtained after alcohol sink classification and named SCP-30,SCP-50 ? SCP-80,respectively.Anticoagulant experiments in vivo and vitro showed that polysaccharides and its different fractions of sweet corn cobs have a significant impact on bleeding time and blood coagulation time,all the components can prolong APTT and TT,but have no significant effect on PT,The anticoagulant effect of SCP-80 was better.
