Sweet corn cob as raw material,the use of solvent extraction and microwave-assisted extraction of phenolic active substances which were extracted using resin polyphenol extracts from purified Extraction and purification of the optimal process conditions.Hyperoside in sweet corn cob polyphenols as a template molecule produced using precipitation polymerization imprinted polymer,formulation optimization,matching and preparation conditions,and to analyze the adsorption characteristics and molecular recognition of molecularly imprinted polymers inspection.The main results obtained are as follows:(1)Sweet corn cob determine the extraction of polyphenols in optimal conditions.In this study,the single factor experiment on sweet corn cob extract polyphenols greatest impact on four factors(ethanol concentration,extraction temperature,extraction time and solid-liquid ratio),orthogonal experiment and response surface experiments established that mathematical model of four factors and the extraction rate.Optimum process parameters obtained by solvent extraction Extraction of sweet corn cob polyphenols as follows:ethanol concentration 70%,extraction temperature was 80?,extraction time was 2.5 hours,solid-liquid ratio of 1:25g/mL and extracted for 3 times,under this condition,get sweet corn cob polyphenol extraction rate of 81.23%.Optimum parameters using microwave-assisted Extraction of sweet corn cob polyphenols as follows:70.77%ethanol concentration,extraction temperature 53.87?,extraction time 2.83min,solid-liquid ratio was 1:38.77 g/mL.Sweet corn cob polyphenol extraction under these conditions,the extraction rate was 94.4%.(2)Static adsorption and desorption experiments,selected DA-201 resin Sweet corn cob purified polyphenols best.Optimal conditions for purification by dynamic adsorption experiment results are as follows:the sample liquid polyphenol concentration of 2.5mg/mL,the sample flow rate of 2mL/min,the sample pH value of 6.85,the sample volume 4BV.Dynamic elution experiments for the optimum elution conditions:eluant concentration(ethanol)70%,eluant rate 3mL/min.(3)Preparation Process Hyperoside molecularly imprinted polymer is:Take the template molecule Hyperoside 1mmol ultrasonic assisted dissolved in 70mL tetrahydrofuran,16mmol functional monomers methyl methacrylate and 30mL toluene,ultrasound deoxy 30min,room temperature under polymerization 12h,so that the template molecule and functional monomer fully integrated,cross-linking agent was added 72mmol EDMA,226mg initiator AIBN,the mixture was poured into a flask three double-deoxy ultrasound 10min,nitrogen gas was bubbled 10min,the container sealed nitrogen environment.The three double bottle connection with 60? water bath and magnetic stirring(120r/min),the polymerization 24h.End of the polymerization,a polymer to give a yellow precipitate,color almost clear colorless solution.The resulting polymer was vacuum dried 70 deg.]C,the resulting yellow powder was treated with acetic acid-methanol:extracting(19)until the solution was subjected to Soxhlet extraction was no change in UV absorbance value,in order to remove the template molecule,white polymer was obtained,and then washed with methanol to acetic acid,which was placed in a blast oven dried 60? 24h,to give a white powdery polymer,namely molecularly imprinted polymers(MIPs).
