Radix Sophorae subprostrata,belongs to genus Sophora subprostrata is a plant distributed mainly in the Limestone Mountain of Guangxi province.It has been used in traditional chinese medicine as an antipyretic,analgesic,alexipharmic,and antiinflammatory agent to treat diseases such as sore throats,jaundice,and cough.Endophytes are widely recognized as prolific sources of biologically active and structurally unique secondary metabolites.Xylaria sp.is a genus of ascomycetes commonly found as wood-decaying and endophytic fungi.The genus Xylaria sp.was found to produce rich secondary metabolites.This article made an investigation on the secondary metabolites from a fungus xylaria sp.Obtained from the Radix Sophorae Subprostrata to get new compounds.This article consisted of two parts.The first part introduced the recent research progress on metabolites from the fungus xylaria sp.The second part includes isolation and identification of the endophytic fungi from the Radix Sophorae Subprostrata,and the isolation,structure identification of secondary metabolites of GDG102.A fungus Xylaria sp.(GDG102)was cultivated in potato glucose liquid medium.Thirteen compounds were isolated from the metabolites of GDG102 through recrystallization,silic gel column,C18 gel column,Sephadex LH-20 columnchromatograph and HPLC methods.Eleven structures of them were established through NMR and MS methods,singlecrystal X-ray diffraction analysis,and Marfey's method.Their structures were elucidated as cupressolide A(1)?1,1 Oa,13-trihydroxyeremophil7(11)-en-12,8-olide(2)?mairetolide B(3)?1 a,1 Oa-epoxy-13-hydroxyeremophil-7(11)-en-12,8?-olide(4)?mairetolide G(5)?xylareremophil(6),3-methyl-7-carboxy8-hydroxyisocoumarin(7)?diaporthin(8)?xylapeptide A(9)?xylapeptide B(10)?xylapeptide C(11).Compounds 3 and 5 were isolated from microorganism for the first time,while 6 was isolated as a new eremophilane sesquiterpene,and 9-11 were new cyclic pentapeptides.The planar configurations of 9 and 10 were elucidated on the basis of spectroscopic methods,and the absolute configurations of all amino acids were determined by Marfey's method and singlecrystal X-ray diffraction analysis.
