The reflux extraction process and the purification process with macroporous resin of Sophorae Flavescentis Radix-Glycyrrhiza Uralensis were investigated based on quality by design(QbD)in this work.The main work is summarized as follows:1.Optimization of Sophorae Flavescentis Radix-Glycyrrhiza Uralensis extraction process following the QbD.The extraction ratios of total alkaloids of Sophorae flavescens(TASF),matrine(MT)and oxymatrine(OMT)were identified as the critical process assess indexs.Single factor experiments and screening tests of significant factors(Plackeet-Burmann design)were applied to identify critical process parameters(CPPs).The central composite designed experiments were performed to build model between CPPs and process assess indexs.The recommended operation space of parameters was as follow:based on Sophorae Flavescentis Radix mass of 7.50 g,Glycyrrhizauralensis mass of 10.0~10.5g,ethanol concentration of 65%~68% and extraction temperature of 94.5?~95.0?.The verification experimental results agreed well with prediction results,which means that model is predictive and operation space is reliable.2.Optimization of extract of Sophorae Flavescentis Radix-Glycyrrhiza Uralensis purification process following the QbD concept.First,NK-109 macroporous resin was identified for further investigation.3BV and 6BV of sample solution and eluent solution were determined by single factor experiments.The desorption mass of TASF,MT and OMT were identified as process assess indexs.Plackeet-Burmann design was applied to identify CPPs.The central composite designed experiments were performed to build model between CPPs and process assess indexs.The recommended operation space of parameters was as follow: sample solution pH of 5.5~6.0,elution ethanol concentration of 83%~84%,elution rate ?2.5BV/h.The verification experimental results agreed well with prediction results,which means that model is predictive and operation space is reliable.3.Draw the growth curve of Escherichia coli and Staphylococcus aureus.Inhibition zone and minimum inhibitory concentration of 65% ethanol extract(A),petroleum ether extract(B),chloroform extract(C),n-butanol extract(D)and purification with NK-109 resin(E)were determined.The results showed that the inhibiton of various substances on Escherichia coli from strong to weak is: E> C> D> A> B;inhibition of Staphylococcus aureus from strong to weak is: E> C > A> D > B.The results showed that A,B,C,D,E have different bacteriostasis on Escherichia coli and Staphylococcus aureus.
