| Actinomycetes have many metabolites,especially antibiotic metabolite used in the treatment of disease and concern,in this master's degree thesis,aims are isolation and identification of actinomycetes from different habitats,the preliminary antibacterial activity and cytotoxicity of the metabolites from actinomycetes which have been screened were studied.In the master's degree thesis,the appropriate method of screening and isolation medium including 50 ug/ml potassium dichromate as bacteria,fungi inhibitors were used in different ecological environment(soil,oceans),34 actinomycetes were obtained.Experiment by agar diffusion method to get the confrontation actinomycetes preliminary screening,and ultimately selected three actinomycetes have some antibacterial activity,molecular biology by 16 S r DNA sequencing of isolated part actinomycetes sequencing,the sequencing results on the network NCBI BLAST sequence alignment showed that screened this three actinomycetes belong to Nocardia asteroides X1,Micromonospora inyonensis X2 and Micromonospora agamiensis X3.A strains which have good activity belonging to Nocardia genus and asteroids species named Nocardia asteroids X1.The antibacterial effect on the extracellular and intracellular antimicrobial substances from Nocardia asteroids X1 were studied and the results show that Nocardia asteroids X1 extracellular and intracellular substances have certain inhibitory effect on Staphylococous,Bacillus and Candia Albicans.In additional,the antimicrobial activity of Actinomycetes against pathogens was determined by the agar block method experiments on pathogenic bacteria,the use of another measuring actinomycetes crude fermentation product with anti-tumor activity for CCK-8 cell A549 method,the results of three have some antibacterial activity.Finally,strains which have good activity measured in the experiment had been deposited with Collection glycerol preservation method in Zhejiang Hisun Pharmaceutical Co.,Ltd. |
