Gynura procumbens(Lour.)Merr(GPM)is a medicinal plants with some functions,such as anti-inflammatory,antihypertensive and hypoglycemic function.Chlorogenic acid and its dimer are the most significant functional components in GPM.In this study,the extraction,purification and in vitro digestion technological parameter of CA dimer from GPM were studied significantly.Also,the antioxidation of digestive product and metabolism in rats were investigated.In addition,the solid beverage of GPM was developed.The results were showed as follow:1.Chlorogenic acid dimer was extracted from Gynura procumbens by ultrasonic-assited method,and the effects of ethanol concentration,liquid to solid ratio,ultrasonic time and ultrasonic power on the yield of chlorogenic acid dimer were studied by Box-Behnken design and response surface methodology.The optimized extraction conditions for maximum yield(35.3mg/CAg DW)of total chlorogenic acid dimer from Gynura procumbens were as followed:ratio of liquid to solid(17m L/g),ethanol concentration(55%),ultrasonic power(450w),ultrasonic time(18min),and extracting two times.2.Optimization of purification technology of chlorogenic acid dimer from Gynura procumbens static adsorption and desorption test was used to select the optimal resin,and dynamic adsorption and desorption conditions were optimized by single factor tests.The results show that the most suitable resin for purification of chlorogenic acid is AB-8 with high absorption and desorption rate.The optimal resin adsorption conditions were 2m L/min of sample flow rate;2mg/m L of sample concentration;p H 3;5BV of sample volume and the optimal desorption conditions were 2m L/min of eluting speed;2BV of eluting volume;30% of ethanol.The purity and yield of chlorogenic acid could be up to 76.5% and 58.4% respectively after purification..In addition,the chlorogenic acid dimer of Gynura procumbens were identified by HPLC-QTOF-MS/MS preliminarily.3.The chlorogenic acid dimer(76.5% of purification)was monitored at in-vitro simulated oral,gastric and intestinal level and the antioxidant activity of digestiveproduct was detected respectively.Results showed that the digestive product during intestinal digestion had the strongest antioxidant activity,following the digestive product during gastric digestion and the oral digestion is the worst.Further study showed that some of chlorogenic acid dimer resolved into caffeic acid and quinic acid and the digestive product during intestinal digestion showed the most caffeic acid and quinic acid which showed stronger antioxidant activity than chlorogenic acid dimer itself.4.The plasma and urine metabolites of rat after oral administration were identified and quantified by UPLC-ESI-QTOF-MS/MS.2 original compounds and 9new metabolites were detected in plasma,while 27 new metabolites and 2 original compounds were observed in urine.The metabolites were mainly compounds with glucoside and sulfo group.5.The G.procumbens solid beverage was developed and its color-protecting,spray drying conditions were optimized.The results showed that the optimal color-protecting conditions were 95? of blanching temperature,75mg/L of zinc gluconate and 6 of PH,20 s of blanching time and 0.4% of Vc.The optimized hydrotropied conditions were 30% of maltodextrin,1% CMC and 1% of2,3-Dihydroxypropyl octadecanoate.The dry-spraying conditions were 180? of inlet temperature,95? of outlet temperature and 30 m L/min of sample speed.Under these conditions,the G.procumbens solid beverage had a relatively good sensory and instant quality.
