As the origin of Prunus mume,China has a very rich resource of it,but the use of Prunus mume in China is mainly concentrated on the food industry.The use of non-fruit parts such as plum flower is relatively lacking,resulting in waste of resources in the annual flowering period.The study found that Prunus mume and Prunus mume flower have a variety of physiological effects and pharmacological effects,the most prominent of which is its antioxidant effect.In order to further study the anti-oxidant effect of Prunus mume,the total flavonoids were obtained by supercritical extraction of the active constituents of Prunus mume,and the flavonoids were analyzed by high performance liquid chromatography-time of flight high resolution mass spectrometry.The model of cell oxidative damage was induced by hydrogen peroxide stimulation.The antioxidant effects of flavonoids from plum blossoms were verified by a series of experiments.It was proved that the flavonoids of green plum can effectively protect against oxidative damage caused by H2O2,and it can also slow down the oxidative stress response of cells.The experimental results are as follows:1.Extraction of flavonoids from Prunus mume flower:CO2supercritical extraction can quickly and efficiently extract total flavonoids from plum blossoms.The extraction conditions are:feeding Prunus mume flower powder 200 g,extraction pressure 20 Mpa,separation pressure5Mpa,the extraction temperature and separation temperature both were 50°C,and 150 m L of95%ethanol was added as an entrainer at a flow rate of 10 m L/min,and the extraction time was2h.The total flavonoids content in the extract obtained by this method was 91 mg/g.High performance liquid chromatography-time-of-flight high-resolution mass spectrometry was used to qualitatively analyze 20 flavonoids such as catechin and rhamnosin.2.Antioxidant ability of flavonoids from Prunus mume blossoms:DPPH free radical scavenging experiment(VC control)showed that flavonoids of Prunus mume flower can effectively scavenge free radicals in a certain concentration range,and have certain anti-oxidation effect,which is equivalent to the antioxidant capacity of VC.3.The stimulating test of flavonoids from Prunus mume:The red blood cell hemolysis experiment showed that the flavonoid concentration of Prunus mume flower was less irritating to cells under 750?g/m L,and almost no hemolysis reaction occurred,which could be used in subsequent cell experiments.4.Protective effect of flavonoids on oxidative damage of cells:The cell induced senescence damage cell model was stimulated with 800?M hydrogen peroxide.The working concentration of the flavonoid preprotected cells was 100?g/m L in the low concentration group and 500?g/m L in the high concentration group.We found that Prunus mume flavonoids have a good pre-protective effect on H2O2-induced oxidative damage,which can effectively alleviate H2O2inhibition of cell proliferation and reduce apoptosis.At the same time,Prunus mume flavonoids can significantly increase the activity of antioxidant enzymes of SOD and GSH-Px,down-regulate the expression of MMP-1 and IL-1?,and up-regulate the expression of FN and TGF-?1.The above results show that the Prunus mume blossom is rich in flavonoids,and the extract has certain anti-oxidation effect,and can protect the cells from oxidative damage,and also can achieve cell antioxidant and anti-aging features.This antioxidant function is achieved by modulating the amount of intracellular antioxidant enzymes,regulating the secretion of extracellular matrix,and regulating the secretion of inflammatory factors to slow down the inflammatory response.We suspect that Prunus mume flavonoids can effectively alleviate the aging and oxidative damage of human skin,which also provides ideas and theoretical basis for the application of citrus flavonoids in the cosmetics industry as an active ingredient in cosmetics and skin care products.
