Blueberry(Semen Trigonellae),also known as cowberry,blue berry,red bean fruit,etc.,is a perennial deciduous or evergreen fruit tree of the rhododendron family.Blueberry fruit contains a large number of flavonoids,anthocyanins,phenolic acids and other bioactive substances,which have many functions,such as preventing brain nerve aging,anti-inflammation,anti-oxidation,liver protection,weight loss,improving body immunity,and so on.In order to explore the immunomodulatory activity and mechanism of blueberry anthocyanins,the immune activity model of mouse spleen lymphocytes was established.Mouse spleen lymphocytes were aseptically isolated and lymphocyte suspensions were prepared.To provide experimental basis for the development of blueberry anthocyanin functional food,the immunomodulatory activity of blueberry anthocyanins was studied in this experiment when the blueberry anthocyanins with the final concentration of 4,8,16,32,64 mg/ml acted on lymphocytes cultured in vitro(48 h).The experimental results are as follows:1.The safe concentration range of blueberry anthocyanin to spleen lymphocytes in mice was4-64 mg/m L.2.Within the dose range of 4-64 mg/m L,blueberry anthocyanins can promote the proliferation of spleen lymphocytes,the secretion of cytokines IL-6 and TNF-?and the expression of m RNA,suggesting that blueberry anthocyanins play an immunomodulatory function by promoting the secretion of cytokines.3.Blueberry anthocyanin can promote the expression of CD4~+and CD8~+single positive phenotype of T lymphocytes in mouse spleen,and play an immune enhancement role by increasing the proportion of CD4~+and CD8~+single positive T cell subsets.4.Blueberry anthocyanin can increase and activate immune effector lymphocytes by promoting mouse spleen lymphocytes to enter DNA synthesis phase,namely S phase,so as to exert immune enhancement effect.5.Blueberry anthocyanins exert immunomodulatory activity by activating c AMP/PKA signal pathway of spleen lymphocytes.
