Extraction Of Pomelo And Grapefruit Peel Essential Oils And Study On Their Bioactivities

Citrus essential oil is currently one of the most widely used essential oils.According to different varieties,it can be classified into orange essential oil,pomelo essential oil,orange essential oil,etc.According to different organs,it can be classified into leaf essential oil,peel essential oil and flower essential oil.Pomelo peel oil is a natural active substance extracted from peel oil cell layer with various functional activities such as bacteriostasis,antioxidation,anticancer,etc.,and its development and utilization have attracted much attention.In this paper,three kinds of grapefruit peel essential oil were extracted by hydro distillation(HD).The chemical composition of the essential oil was determined by gas chromatography-mass spectrometry(GC-MS),and the corresponding antibacterial,antioxidant and anticancer activities were determined.Cold-pressed grapefruit essential oil was used as a raw material,and the corresponding biological activity experiment was performed on the separated light-phase components through molecular distillation technology.The main findings were as follows:1.Three kinds of pomelo peel essential oils were extracted by HD,and the chemical composition of the peel essential oil was determined by GC-MS.11,15 and 20 components were identified in Shatian pomelo,red meat honey pomelo,and three red honey pomelo,respectively.Their main ingredients were limonene and myrcene.The filter paper method preliminarily evaluated the bacteriostatic activity of three HD extracted essential oils against four common bacteria.The bacteriostatic test results showed that the red meat honey pomelo had the best inhibitory effect on Escherichia coli and the worst on Bacillus subtilis.Sanhong honey pomelo has the best inhibitory effect on Staphylococcus aureus,and the worst on Bacillus subtilis.Shatian pomelo was not sensitive to all four bacteria.The anticancer test results showed that when Shatian pomelo essential oil concentration was 5?L/m L,the essential oil had a significant inhibitory effect on the proliferation of A549 lung cancer cells,and the half inhibitory concentration(IC₅₀)of A549 lung cancer cells was 2.97?L/m L.2.Cold-pressed grapefruit essential oil was prepared by molecular distillation.GC-MS was used to determine the chemical composition,and light-phase components were used for antibacterial,antioxidant,and anticancer biological activity experiments.A total of 24 compounds were identified,and limonene was the main component,accounting for 93.33%of the total oil.The bacteriostatic test results showed that grapefruit essential oil had the strongest inhibitory effect on Bacillus subtilis,followed by E.coli,Staphylococcus aureus,and Salmonella.Grapefruit essential oil had no inhibitory activity against Pseudomonas aeruginosa.The antioxidant test results showed that the IC₅₀ of grapefruit light phase scavenging DPPH and ABTS free radicals were 22.06 mg/m L and 15.72 mg/m L,respectively.The results of the anticancer test showed that when the concentration of the essential oil was 0.3?L/m L,the inhibition of the proliferation of both cancer cells was significant.When the concentration of essential oil was 0.6?L/m L or higher,the survival rates of Hep G2 liver cancer cells and HCT116 colon cancer cells were both lower than 15%.When the grapefruit peel essential oil concentration was 0.3?L/m L,it had a significant inhibitory effect on the migration of cancer cells.The IC₅₀ values of Hep G2 and HCT116 were0.24?L/m L and 0.20?L/m L,respectively.In short,pomelo and grapefruit peel essential oils are bioactive natural products.The research on their development and utilization will not only help to improve the industry chain and enhance the commercial value,but also benefit for the identification and quality evaluation of pomelo and grapefruit and their use in food.Our research will provide scientific reference and theoretical basis for the development of antibacterial preservatives and anticancer drugs...
