Molecular imprinting technology(MIT)is an interdisciplinary technology derived from the disciplines of material chemistry,polymer chemistry,biochemistry,etc.,which has a similar mechanism of immunology as"antigen-antibody".The molecularly imprinted polymer(MIP)prepared by molecular imprinting technology has the advantages of specific recognition,selective adsorption of target compounds and good chemical stability.It has been widely used in solid phase extraction,capillary electrophoresis,liquid chromatography,sensors,and other fields.Quercetin in flavonoids is widely present in flowers,fruits,and leaves of plants.It has a variety of pharmacological effects and biological activities.It is often used to prevent and treat tumors and cardiovascular diseases.Bolete is a delicious and nutritious wild edible mushroom,It contains a large amount of antioxidant glutathione,which is much higher than other wild edible fungi,and has high edible value and medicinal effect.In this paper,core-shell nanoparticles with superparamagnetism are used as carriers,quercetin and glutathione as template molecules,respectively,and new magnetic molecularly imprinted polymer(MMIPs)with specific recognition and selective adsorption of template molecules were prepared by surface molecular imprinting technique.Magnetic molecularly imprinted polymers(MMIPs)were characterized by Fourier transform infrared spectrometer(FT-IR),X-ray diffraction(XRD),transmission electron microscopy(TEM)and vibrating sample magnetometer(VSM),and their adsorption properties for template molecules were studied.The main research contents and results of this paper are as follows:1.Magnetic molecularly imprinted polymer combined with high performance liquid chromatography for selective extraction and determination of the metabolic content of quercetin in rat plasmaIn this study,a novel magnetic solid phase extraction adsorbent was prepared,namely Fe3O4@Si O2 as the carrier,quercetin as the template molecule,acrylamide(AM)as the functional monomer,ethylene glycol dimethacrylamide(EGDMA)as the crosslinker,2,2'-azobisisobutyronitrile(AIBN)as the initiator to prepare magnetic molecularly imprinted polymer(MMIPs)with specific adsorption of quercetin by surface molecular imprinting technique.MMIPs were characterized by Fourier transform infrared spectrometer(FT-IR),etc..The binding properties of MMIPs were evaluated by isothermal adsorption,kinetic adsorption and selective adsorption experiments.The results showed that the saturated adsorption capacity and adsorption time were 17.9?mol/g and 20 min,respectively,and showed high selectivity for quercetin.Under optimized conditions,the limit of detection(LOD)and limit of quantitation(LOQ)were 55.327 ng/m L and 184.413 ng/m L,respectively.The recovery and RSD were 85.34?95.67%and 3.36?6.59,respectively.We successfully combined magnetic solid phase extraction with high performance liquid chromatography(MSPE-HPLC-UV)for the enrichment,separation and detection of quercetin in rat plasma.The results showed that the peak and half-life of quercetin in rat plasma were 1.5 h(32.310?g/m L)and 3 h,respectively.The method was simple,rapid and efficient,and could be applied to the separation and detection of quercetin in complex matrices,and also provides a basis for the separation and identification of other natural chemical components.2.Magnetic molecularly imprinted polymer combined with high performance liquid chromatography for the selective separation and determination of glutathione in various wild edible boleteIn this study,a novel magnetic molecularly imprinted polymers(MMIPs)with specific adsorption for glutathione were prepared.Based on magnetic molecularly imprinted polymers(MMIPs)as adsorbents for magnetic solid phase extraction,we combined magnetic solid phase extraction with high performance liquid chromatography(MPSE-HPLC)for the separation,enrichment and detection of glutathione in nine kinds of wild edible bolete extracts.Magnetic molecularly imprinted polymers(MMIPs)were characterized by Fourier transform infrared spectrometer(FT-IR),etc..In addition,the properties of magnetic molecularly imprinted polymers(MMIPs)were studied by adsorption isotherm,adsorption kinetic and selective adsorption experiments.The results showed that MMIPs had high selectivity for glutathione(kd=245.33 m L/g),adsorption time and saturated adsorption capacity were 30 min and 17.88?mol/g,respectively.Under the optimized conditions,there was a good linear relationship in the concentration range of 100?3000?g/L,the correlation coefficient R2=0.9997,the limit of detection(LOD)and the limit of quantitation(LOQ)were 24.828?g/L and 82.760?g/L,respectively.The spiked recovery and RSD were 93.28?103.03%and 2.77?5.11,respectively.We successfully combined magnetic solid phase extraction with high performance liquid chromatography(MSPE-HPLC)for the detection of glutathione in nine kinds of wild edible bolete extracts.It was found that the content of glutathion in Leccinum extremiorientale(L.Vass.)Sing.was the highest(19.800 mg/g)and that in Boletus aereus was the lowest(6.791 mg/g).
