With the increasing severity of metabolic problems,including obesity and diabetes,consumers' concept of healthy life and awareness of low sugar are gradually improving.Therefore,artificial sweeteners are widely used as food additives because of their high sweetness and low energy.The application of sucralose is more popular,but its safety evaluation is still at the level of toxicity,and its potential health effects on human intestinal microecology and intestinal immunity should also be included in the safety evaluation system.Therefore,this study aims to explore the effects of different concentrations of sucralose in mice,and explore the effects of sucralose on the intestinal microecology of mice from the physiological indexes,glucose and lipid metabolism and intestinal barrier integrity.Combined with the above indicators to speculate whether sucralose may cause inflammatory bowel disease and insulin resistance and gradually induce type 2 diabetes.This study provides a research basis for the mechanism of adverse effects of sucralose on human health,and provides guidance and new ideas for the practical application of artificial sweeteners.In this study,BALB / c mice were randomly divided into five groups: blank control group(purified water),high concentration sucralose group(3000 mg / kg),medium concentration sucralose group(1500 mg / kg),low concentration sucralose group(15 mg / kg)and high concentration sucralose positive control group(3000 mg / kg).After 6 weeks of feeding,the mice were killed,and the serum,spleen,thymus,spleen,spleen and spleen were taken To explore the effects of sucralose on the basic physiological indexes(body weight change,impaired fasting blood glucose,glucose tolerance,blood lipid),intestinal biological barrier(intestinal flora structure),intestinal chemical barrier(short chain fatty acids,total metabolites),intestinal physical barrier(intestinal microstructure,total metabolites)of mice Intestinal villus length and crypt depth),intestinal immune barrier(cytokines IL-1 ?,IL-6,TNF – ?,CRP,LPS,SIg A).The results showed that in terms of physiological indexes,the long-term intake of sucralose could lead to the growth of mice,the body tended to be impaired fasting blood glucose and glucose tolerance;In terms of intestinal biological barrier,the diversity and abundance of intestinal flora are reduced,the structure of intestinal flora is changed,and the proportion of beneficial bacteria and harmful bacteria is unbalanced,which leads to the damage of intestinal biological barrier;The intestinal barrier will cause the secretion of intestinal metabolites to be disordered,the content of SCFAs and the expression of the metabolites in the characteristic feces are significantly changed,and the intestinal chemical barrier is destroyed;In terms of physical barrier,the pathological microstructure showed that the intestinal epithelium was damaged,inflammatory cells gathered,fibrous connective tissue proliferated,digestion and absorption function weakened,and physical barrier was damaged;On the basis of the damage of three intestinal barriers,cytokines IL-1 ?,IL-6,TNF-?? High expression of CRP,LPS was injected into blood,SIg A content decreased,and intestinal immune barrier was damaged.The results showed that the intestinal barrier of mice was damaged,and the microecological balance of intestine was broken,which led to intestinal inflammation in mice.Therefore,insulin resistance and type 2 diabetes mellitus were induced in mice.In conclusion,long-term intake of sucralose can greatly increase the possibility of insulin resistance and type 2 diabetes by affecting the growth of the body,destroying the integrity of intestinal mucosal barrier,affecting the intestinal microecological balance,and up regulating intestinal inflammation.Low concentration of sucralose in accordance with ADI standard can also cause changes in intestinal flora structure,flora abundance and characteristic metabolites to a certain extent,selectively express some characteristic metabolic pathways,and increase the expression of some inflammatory cytokines.
