Acrylamide(AA)is a moderately toxic substance with neurotoxicity,genotoxicity,reproductive toxicity and potential carcinogenicity,and it is widely present in the daily diet.Therefore,the prevention and control of acrylamide toxicity has become a worldwide research hotspot.Studies have confirmed that oxidative stress is the main mechanism of acrylamide-induced toxicity in vivo.Therefore,inhibiting acrylamide-induced oxidative stress is an important way to prevent and control its toxicity.As one of the plants with a long history in my country,seabuckthorn can be used for medicine and food.Its fruit is rich in a variety of active substances,such as vitamins,flavonoids,superoxide dismutase,etc.,and has excellent antioxidant effects.However,it is not clear whether seabuckthorn berry juice can effectively alleviate the oxidative damage induced by acrylamide in vivo.Therefore,this study established a subacute acrylamide exposure rat model on the basis of determining the antioxidant components and antioxidant properties of seabuckthorn berry juice to explore the mitigation effect of seabuckthorn berry juice on acrylamide-induced toxicity in rats.The specific research content is as follows:1.Determination of antioxidant components and antioxidant capacity of seabuckthorn berry juice(1)Determine the content of vitamin C,total phenols and total flavonoids in sea buckthorn juice,and compare the antioxidant capacity of seabuckthorn berry juice with different concentrations(2%,4%,6%,8%,10%,20%,40%,50%),Further explore the difference in antioxidant capacity between 20% concentration of seabuckthorn berry juice and a single component.The results showed that the vitamin C content in seabuckthorn berry juice was 135.65mg/100 m L,the total phenol content was 138.38 mg/100 m L,and the total flavonoid content was70.79 mg/100 m L;the scavenging rate of seabuckthorn berry juice on three free radicals varied with the concentration.The increase in the juice is constantly increasing,and the increasing trend gradually flattens when the juice concentration is 20%.When the juice concentration is 50%,the three free radicals have the highest scavenging rate.At this time,the scavenging rates of superoxide anion free radicals,hydroxyl free radicals and DPPH free radicals are 85.39%,92.03%,and 96.77% respectively;compared to other single components seabuckthorn berry juice has the highest scavenging rate for these three free radicals,followed by vitamin C,total phenols,and total flavonoids,and there are significant differences between the groups(p <0.05).2.Research on the protective effect of seabuckthorn berry juice on the toxicity in rats(1)Establishment of animal model: This experiment uses male SD rats as animal models and randomly divides them into four groups.The blank control group,the poisoned group(acrylamide),the positive control group(vitamin C),and the seabuckthorn berry juice intervention group.The drug was administered by oral gavage,and the test period was 15 days.(2)Explore the effect of seabuckthorn berry juice on the overall health of rats by measuring the changes in body weight of rats during the test.The results showed that the weight of the rats in the four groups kept increasing steadily during the experiment,and the growth rate of the rats in the control group was the highest.At the end of the experiment,the weight growth rate of the rats in the seabuckthorn berry juice intervention group was significantly higher than that of the rats in the exposure group,but lower than the rats in the control group.(3)Explore the protective effect of seabuckthorn berry juice on neurotoxicity of rats by measuring the neurobehavioral indexes of gait score and hind limb splay.The results showed that the hind limbs of the rats in the poisoned group were dragged and valgus,the gait was obviously abnormal,and the hind limb supporting force was significantly weakened.Although the rats in the seabuckthorn berry juice intervention group had slight gait abnormalities,compared with the rats in the poisoned group,the hind limb supporting force was significantly increased,and the ataxia was alleviated.(4)Explore the protective effects of seabuckthorn berry juice on the blood protein level,blood lipid level and liver and kidney function of rats by measuring the blood biochemical indicators of rats.The results showed that the total protein and albumin content of rats in the seabuckthorn berry juice intervention group did not change significantly;total cholesterol,triglyceride,creatinine,urea nitrogen content,lactate dehydrogenase,alanine aminotransferase,aspartate aminotransferase and alkaline phosphatase activities significantly lower than the poisoned group.It shows that seabuckthorn berry juice has no obvious effect on the blood protein level of rats,but it can effectively improve the increased permeability of liver and kidney cell membranes in rats caused by acrylamide.(5)To evaluate the repair effect of seabuckthorn berry juice on the oxidative damage of rat brain hippocampus,cerebellum,liver,small intestine and kidney tissues.The results show that the intervention of seabuckthorn berry juice can significantly increase the activity of superoxide dismutase,catalase and the content of glutathione in various tissues and organs;reduce the content of malondialdehyde in various tissues and organs.(6)Hematoxylin-eosin staining was performed on the hippocampus,cerebellum,liver,small intestine and kidney of rats,and the pathological effects of acrylamide on the above-mentioned tissues were observed under an optical microscope.Pathological observations show that seabuckthorn berry juice can alleviate acrylamide-induced neuronal necrosis in the hippocampus of the brain,necrosis of Purkinje cells in the cerebellum,decrease in the number of granular cells,rupture of the small intestine villi,unstriped arrangement of liver cells and glomerulus atrophy and other injuries.In summary,subacute exposure to acrylamide can cause oxidative stress in various tissues and organs of rats,which in turn leads to slow weight gain,nervous system damage,and damage to the structure and function of various organs.Seabuckthorn berry juice can effectively improve the ability of the rat antioxidant system to resist oxidative stress induced by acrylamide through its strong antioxidant properties,and reduce the oxidative damage of various tissues and organs caused by lipid peroxides.Seabuckthorn berry juice has achieved the protective effect of acrylamide-induced toxicity in rats.
