Objective:Yizhi chewable tablets is a kind of health food with the functions of filling marrow,nourishing blood and tonifying deficiency,tranquilizing and replenishing intelligence,which is based on Liuwei Dihuang pills.It provides experimental basis for the further development of Yizhi chewable tablets.In this study,the prescription compatibility,preparation technology,functional evaluation,safety evaluation and quality standard of Yizhi chewable tablets were systematically studied.Method : Through literature research,the components of each drug and the mechanism of related drugs were analyzed to test the rationality of the thinking of the prescription;meanwhile,functional experiments were used to optimize the prescription dosage and orthogonal experiment was used to optimize the technology;scopolamine model mice were tested by platform jumping experiment,dark avoiding experiment and Morris water maze experiment Prescription improves memory function.In addition,the potential target of Yizhi chewing tablets to improve memory impairment was predicted and screened by using the network pharmacology research technology and molecular docking technology,which provided theoretical basis for its potential mechanism.On this basis,the combination characteristics of prescription of Yizhi chewable tablets were further explained by using molecular docking technology to verify the reliability of target prediction.After the pretreatment of the raw materials in the prescription,orthogonal design test was adopted in the extraction process.The content of total saponins was investigated.The soaking time,the time of decoction,the amount of water added and the number of times of decoction were four factors.Through the design of orthogonal test,the process parameters of water extraction were determined,and the best water extraction process was selected and verified.In the forming process,water extract is the main raw material.When choosing the appropriate auxiliary materials,the filler,adhesive and flavor straightener are optimized and selected in the amount of use by single factor test method.The optimal scheme is determined.The preparation method is simple,sweet and delicious,and the edible and easy to carry.The process validation is carried out according to the optimal process.After 30 days of continuous gavage of ICR mice,the mice in other groups except the blank group were given 5 mg/kg scopolamine 5 mg/kg by intraperitoneal injection,and each group was injected with scopolamine 5 mg/kg,so as to prepare the memory impairment model.After modeling,the changes of learning and memory ability of mice in each group were further evaluated by jumping off the platform experiment,dark avoiding shows that the main active components of Yizhi chewable tablets are through the interaction pathway of active ligand receptor,vascular endothelial growth factor signal pathway and cholinergic synaptic signal pathway.The Yizhi chewing tablets may be regulated by proliferation and apoptosis,inflammatory response and inflammatory response through the active component cells such as cousterol,?-glutasterol,diosgenin,and haifengtengone AKT1,VEGFA,PTGS2,ACh,ache,and chat target play an important role in improving memory.The best preparation technology of Yizhi chewing tablets is: soak the raw materials in water for 1 hour,add to 10 times the volume of water,cook twice,boil for60 min each time,filter them,concentrate into extract and get the extract powder by decompression compatibility of Yizhi chewable tablets was comprehensively elaborated,the process parameters were determined,the function and safety of Yizhi chewable tablets in improving memory impairment were evaluated,the quality standards were formulated,and the development of Yizhi chewable tablets was generally completed,which is worthy of further promotion and application.
