Constipation and diabetes are common pregnancy complications affecting the pregnancy outcome to a certain extent for overweight and obese pregnant women,meanwhile less drugs are available to pregnant women.Many researches had been proved that the probistics could affect intestinal microecology which could influence constipation and diabetes.In this study,based on the clinical trials it had been demostrated that probiotics fermented milk could effectively regulate the intestinal microecology for overweight and obese pregnant women inorder to improve constipation and reduce the diabetes during pregnancy..After recruiting volunteers from overweight and obese pregnant women at the first trimester of pregnancy,they were randomly divided into two groups: probiotic-rich fermented milk pre-treatment group and routine control group.Comparing with the control group,the volunteers began to drink a bottle of fermented milk every day from the 13 th week of pregnancy.The faeces samples were collected from the volunteers in the test: first sampling(8-12w),second sampling(24-28w),third sampling(34w).The intestinal microorganisms were detected by the V3-V4 area of 16 Sr RNA.Two group would filled in the self-rating constipation scale at begin of enrolled in the group(12weeks).Then just the group fed with fermented milk filled in the self-rating constipation scale.once a month,for a total of 7 times.Oral glucose tolerance test was performed in both groups at 24-28 weeks.It was statistically analyzed that the constipation scale data,oral glucose tolerance data and sequencing data.The results showed that fermented milk could significantly improve the constipation symptoms with low fecal volume for pregnant women.Compared with the conventional control group,the incidence of gestational diabetes decreased from 32% to 22% and the total number of OTUs in the entire pregnancy was 2646 more than 1398.The contribution rates of the first and second principal coordinates of PCo A weighted analysis are 36.1% and 7.48%respectively,while those of non-weighted are 10.3% and 5.96% respectively.The analysis of Anosim,Adonis and AMOVA showed that there was significant difference between the intervention group and the control group.T-test,Meta Stat,LEf Se and other analysis found differences in species such as Clostridia.Tax4 Fun analysis showed that intestinal flora was related to genetic information and human diseases.The studies had been demonstrated that probiotic-rich fermented milk intake improving intestinal microecological could effectively alleviate the symptoms of constipation and reduce the incidence of diabetes and provided some reference basis for probiotics to improve the healthy pregnancy.
