Objective:At present,the development of polysaccharide,one of the main active components of Cardamine hupingshanensis,is still shallow,and its bioactive function is lack of exploration.In this paper,Cardamine hupingshanensis in Enshi area was selected as the material.Firstly,the effects of ultrasound on the extraction amount,microstructure and color of Cardamine hupingshanensis polysaccharide were explored.Then explore the effect of different doses of Cardamine hupingshanensis polysaccharide(CHP)on the intestinal flora of normal mice and function prediction.Methods: The content of polysaccharide was determined by phenol-sulfuric acid method;the microstructure of polysaccharide was observed by scanning electron microscope;the color of polysaccharide was measured by CS-820 N Desktop Spectrophotometer.16 s rRNA was used to determine intestinal contents;UHPLC-QE-MS was used to determine metabolites in plasma.Results:(1)The ultrasonic extraction kinetic model was established,and the equations with relevant parameters and correlation coefficients above 0.90 were obtained.When the extraction conditions reached 300 W,353K,and 60 min,the maximum extraction amount was 0.7146mg/ml;The microstructure of polysaccharides extracted by ultrasonic and water is different,and the granular shape of polysaccharides extracted by ultrasonic is obvious;Compared with water extraction,ultrasound L~* and ?E were significantly increased(P<0.05),and a~* was significantly reduced(P<0.05).(2)Cardamine hupingshanensis polysaccharide decreased the relative abundance of Firmicutes and increased the relative abundance of Bacteroides,and the ratio of Bacteroides to Firmicutes increased.(3)Polysaccharide and metformin improved blood glucose,CRE,HDL-C,BUN,liver to body weight ratio.After intragastric administration of metformin and polysaccharide,Oscillibacter?Roseburia?Marvinbryantia?Blautia?Candidatus?Arthromitus were observed in type 2 diabetic mice?The content of genus was lower than that of TC group.but in Lactobacillus?Ruminococcaceae?UCG-014?Anaerotruncus?uncultured?bacterium?f?Erysipelotrichaceae?Parabacteroide?Anaerovorax.The contents of erysipelotrichaceae,parabacteroid and anaerovorax were higher than those of TC group,and were closer to those of ZC group.Compared with normal mice,the normal physiological metabolism in plasma of diabetic mice was seriously disturbed,and the composition of metabolites was changed.Metformin and Cardamine hupingshanensis polysaccharide can improve PC(18:2(9Z,12Z)/18:0)?Pyrazine?DG(20:4(5Z,8Z,11 Z,14Z)/18:2(9Z,12Z)/0:0)?Isolinderanolid?Cervonyl carnitin?DG(22:5(7Z,10 Z,13Z,16 Z,19Z)/18:3(6Z,9Z,12Z)/0:0)N-Oleoylethanolamin?in diabetic mice.(4)The difference of intestinal flora and metabolites was correlated between TC group and ZC group.cis-Acetylacrylate in organic acid inhibited peptococcus and Ruminococcaceae?UCG-014.The homoarecoline in alkaloids can inhibit Klebsiella and uncultured?bacterium?f?Rs-E47?termite?group is a growth group.PC(18:1(11Z)/14:0)of Phosphatidylcholines can inhibit the Lactococcus and Eschericia Shigella,which contains many harmful bacteria.PC(20:3(8Z,11 Z,14Z)/14:0)in glycerophosphatids can promote Klebsiella and Ruminococcaceae?UCG-014 genus,inhibited uncultured?bacterium?o?Mollicutes?RF39 genus.PI(18:1(11Z)/18:3(6Z,9Z,12Z))of glycerophosphatides can promote the enterococci and inhibit Candidatus?Soleaferrea.Conclusion:(1)ultrasonic extraction can improve the extraction amount of polysaccharides,change the microstructure of polysaccharides,and is conducive to the removal of pigment.(2)The structure of intestinal flora in normal mice was changed by polysaccharide from Cardamine hupingshanensiss.(3)The polysaccharide of Cardamine hupingshanensis can improve the blood glucose,CRE,HDL-C,bun,liver to body weight ratio,intestinal microorganisms and plasma metabolites of T2 DM mice.
