Dendrocalamus latiflorus Munro is a perennial plant of the genus Phyllostachys in the Gramineae family.There are about 21,300 hectares of Dendrocalamus latiflorus Munro in Chishui.Studies have shown that dietary fiber has the effects of lowering blood sugar,lowering blood fat,anti-oxidation,and preventing constipation.At present,the utilization rate of bamboo leaves is low.Therefore,it is of certain value to improve the comprehensive utilization of bamboo leaves dietary fiber research.In this study,the dietary fiber was extracted from the Dendrocalamus latiflorus Munro leaves by different preparation methods,and the content and functional characteristics of the obtained dietary fiber were measured respectively.Feasibility analysis was conducted on the extraction technology of different preparation methods.The following are the research results of extraction process and Physicochemical characteristics:1.Preparation of dietary fiber of Dendrocalamus latiflorus Munro leaves by alkali extraction method.The single factors of lye extraction,such as,lye concentration,solid-liquid ratio,temperature and extraction time,were optimized by Box-Behnken to obtain the best extraction process.The contents of IDF and SDF were determined.The optimal conditions were as follows:time 90 min,temperature 72 ?,solid-liquid ratio 1:19 g/m L,Na OH concentration 9.5%.Under these conditions,SDF and IDF yields were 39.64% and 51.48%,respectively.The contents were60.52% and 87.94%,respectively.2.On the basis of lye extraction,ultrasonic assisted extraction was adopted.The extraction process was optimized by response surface methodology.The optimal extraction conditions were as follows: time,temperature,solid-liquid ratio and Na OH concentration were 68 min,66 ?,1:19g/m L and 8.5%,respectively.Under these conditions,the average yields of SDF and IDF were45.18% and 46.44%.The contents of SDF and IDF were 64.44% and 89.52%,respectively.3.On the basis of the research group's preliminary experiments and literature research,the extraction process of water extraction method was determined as follows: extraction times of 3times,solid-liquid ratio of 1:12,1:10,1:10 g/m L,extraction temperature of 80 ?/ time,extraction time of 2h/ time;Under these conditions,the average yield of SDF and IDF were 3.42% and85.61%.The contents were 54.05% and 73.45%,respectively.4.According to the method of literature,the water holding capacity,expansion capacity and oil holding capacity of dietary fiber prepared by lye extraction,ultrasonic-assisted lye extraction and hot water extraction were measured.The results showed that there was little difference in the Physicochemical properties of SDF prepared by different methods.Among them,the hydraulic capacity is between 3.23 and 3.59 g/g.The expansive force is between 2.78 and 2.93 m L/g.The oil holding capacity is within the range of 4.05?4.20 g/g.In addition,IDF showed little difference.This paper adopts alkali auxiliary alkali extraction,ultrasonic extraction and water extraction of dietary fiber was studied,and the different methods for preparation of dietary fiber content and Physicochemical properties were determined,the results for the subsequent ingredients like water insoluble fiber preparation of nano cellulose and activated carbon laid a solid foundation,It can also provide theoretical basis and technical support for the comprehensive utilization of bamboo leaf resources in Chishui,and has important theoretical significance and application value for the development and utilization of bamboo resources.
