Sunflowers are widely cultivated in our China,which are easy to obtain,but in agriculture,they are only thrown away as feed,fuel or as waste,resulting in serious environmental pollution and waste of resources.Sunflower disc is one of the main by-products of the sunflower industry.The activity of phenols and other substances have not attracted enough attention.The chlorogenic acid active substances contained in sunflower discs have many important effects:Extracting chlorogenic acid from sunflower discs can not only increase the added value of sunflower discs,but also avoid environmental pollution and resource waste,and has certain economic prospects..The research content and results of the paper are as follows:(1)Extraction of chlorogenic acid from sunflower discIn this study,three methods to extract chlorogenic acid in sunflower trays were compared?Among them,the ultrasonic-assisted extraction method had the highest yield.On the basis of the following five single-factor experiments and optimization by response surface experiments,the optimal technological parameters for extracting chlorogenic acid from sunflower discs were determined as follows:The volume fraction of ethanol is 40%,the ultrasonic temperature is 60?,the material-to-liquid ratio is 1:45g/m L,40 minutes is the ultrasonic time,and the ultrasonic power is 90 W.Under these conditions,the yield of chlorogenic acid in the sunflower disk reaches 6.01 mg/g.(2)Isolation and purification of chlorogenic acid from sunflower discThree macroporous adsorption resins were used to purify sunflower disc chlorogenic acid.The results showed that the AB-8 macroporous adsorption resin had the best separation effect on sunflower disc chlorogenic acid,and the adsorption and desorption were carried out by static and dynamic purification.Experiment,through single factor and orthogonal experiment to determine the best separation and purification conditions.Static adsorption conditions:adjust the p H of the sample solution to 3 before loading the column;dynamic adsorption conditions:sample solution volume is 3 BV,sample solution flow rate is 2m L/min,eluent flow rate is2m L/min,eluent The volume is 4BV,and the yield is 69.48%.(3)Characterization of chlorogenic acid in sunflower discThe qualitative analysis of chlorogenic acid in sunflower disk was carried out by ultraviolet-visible absorption spectrum,infrared spectrum,high performance liquid chromatography and mass spectrometry.The results showed that the characteristic absorption peak of chlorogenic acid in sunflower disk was 330 nm;the main wave number of infrared spectrum was 3252 cm-1,2929 cm-1,1959 cm-1,1379 cm-1,1036cm-1;the retention time of high performance liquid chromatography is 11.818 min,The peak time of chlorogenic acid standard is basically the same;high performance liquid chromatography and mass spectrometry show that the extraction of sunflower disc contains chlorogenic acid substance.(4)In vitro hypoglycemic activity of chlorogenic acid from sunflower discThe a-glucosidase and a-amylase inhibitory activities were used to evaluate the in vitro hypoglycemic activity of chlorogenic acid from sunflower discs.By comparing the different concentrations of chlorogenic acid before purification and after purification,the inhibition rate of different concentrations of a-glucosidase and a-amylase was compared,and acarbose was used as a control to conduct an in vitro hypoglycemic test.The results show that the activities of a-glucosidase and a-amylase will be inhibited by sunflower disc chlorogenic acid to a certain extent,and after purification and at a concentration of 0.8 mg/m L,in addition to acarbose,its effect on a-The inhibition rates of glucosidase and a-amylase can reach 65%and 61%,which are significantly higher than those before purification.Therefore,the results of this study show that sunflower disc chlorogenic acid has a good in vitro hypoglycemic activity.
