| In recent years,the number of non-alcoholic fatty liver patients has increased year by year due to the increase of fat intake,and the age of patients is becoming younger.Unlike long-chain fatty acids,medium chain triglycerde(MCT)has a special structure.The hydrolyzed medium chain fatty acids do not re-synthesize triglycerides for storage as body fat,but are transported directly through the portal vein blood.Use it in the surrounding tissues.Based on this feature,MCT can be quickly digested and absorbed by the human body.In addition to weight loss,rapid energy supply,and improvement of sugar and cholesterol metabolism,MCT’s own emulsifying properties can also promote the digestion and absorption of minerals and some essential fatty acids to a certain extent.However,direct oral administration of MCT will cause adverse reactions such as nausea,vomiting and diarrhea,and that could reduce therapeutic effect of MCT.At present,there are few studies on the effect of MCT on non-alcoholic fatty liver and the preparation of MCT nanoemulsion,and the research on the stability and physical and chemical properties of nanoemulsion is not thorough enough.Therefore,the improvement effect of MCT on non-alcoholic fatty liver is studied in this paper,and high-pressure homogenization technology is used to prepare MCT nanoemulsion.The stability and digestibility of MCT are also studied,which is a good example for MCT in the field of functional food and medicine.First,the improvement effect of MCT on non-alcoholic fatty liver is studied.The results show that: MCT can significantly reduce the body weight,liver weight and liver coefficient of NAFLD mice induced by high-fat diet,and can adjust the four items of blood lipids to make it closer to normal value.Therefore,the metabolism of non-alcoholic fatty liver can be improved by using MCT.In the process of preparing MCT nanoemulsion in the way of high pressure homogenization method,the formula is optimized.The results show that the particle size of the nanoemulsion is affected to a certain extent by the number of homogenization,homogenization pressure,emulsifier mass fraction and oil phase mass fraction.Through 4groups of single factor experiments,the response surface optimization experiment of 3levels and 3 factors is determined.The optimal formula is finally obtained: the mass fraction of emulsifier Tween 80 is 3%,the mass fraction of oil phase MCT is 6%,and the highpressure homogenization pressure is 70 MPa,the number of high-pressure homogenization is 9 times.And the experimental verification shows that the minimum average particle size of the nanoemulsion can reach 185.78±2.33 nm.The type,stability and fatty acid release rate of the prepared MCT nanoemulsion are measured.The results show that: MCT nanoemulsion is an oil-in-water nanoemulsion;after the nanoemulsion has undergone centrifugation,storage,high and low temperature cycles,and temperature stability tests,there is no delamination,demulsification and other phenomena occur,and the particle size changes little,which proves that the prepared nanoemulsion has good stability.The results of transmission electron microscopy show that the nanoemulsion prepared is uniform and spherical.Through the in vitro digestion experiment,it is found that the MCT nanoemulsion has higher fatty acid release rate. |
