Cherry is a wild or semi-wild plant of Rosaceae.It is rich in many kinds of vitamins,anthocyanins,mineral elements,polysaccharides and amino acids.At the same time,it also has certain medicinal value,such as anti-oxidation,anti-inflammatory,anti-freezing injury and so on.At present,there are more reports on the analysis of cherry nutrients,and its deep processing,such as cherry juice,cherry wine and so on,has been developed,and its active components and pharmacological effects have been less studied.The research on cherry residue polysaccharide has not been reported.In order to fully develop and utilize this plant resource,the extraction,purification,structure identification,antioxidant activity and immune activity of sweet cherry lees polysaccharides.The experienment were studied by using sweet cherry less as raw material.The main results are as follows:1.Crude polysaccharides were extracted from the dregs of fruit wine brewed from cherries at 85°C with a material-to-liquid ratio of 1:30 and a water bath for 5 hours.The extraction rate of the crude polysaccharides was6.9%.The removal rate of crude polysaccharide was 83.05%by papain enzyme method and Seveag method.2.Polysaccharides from sweet cherry wine lees were extracted,the soluble polysaccharide fraction was purified on DEAE-52 cellulose and Sephadex G-100 columns to yield a refined polysaccharide YTZ-A.FT-IR,AFM,SEM and UV were applied to elucidate its chemical structure.The molecular weight of YTZ-A was between 4500 and 40000 Da.YTZ-A is mainly composed of arabinose,glucose,mannose,and galactose.The molar ratio of each monosaccharide is 0.97:1.05:0.13:1.17.3.Ascorbic acid was used as positive control to determine the scavenging ability of crude polysaccharide of sweet cherry lees to DPPH free radicals,hydroxyl radicals and superoxide anion radicals.The results showed that anti-oxidation ability of crude polysaccharide from sweet cherry lees is stronger.The IC50value(DPPH)was 0.152 mg/ml,and the IC50value(-OH)was 0.108 mg/ml,and the ability of crude polysaccharide from sweet cherry wine less to remove superoxide anion is weak..4.YTZ-A activation of RAW264.7 macrophages by promoting NO production and TNF-?release has no significant inhibitory effect on cell viability.YTZ-A induce RAW264.7 cells to release NO mainly by activating the JNK MAPK signaling pathway.YTZ-A stimulate the secretion TNF-?.of RAW264.7 cells mainly through NF-?B?JNK MAP?Erk MAPK and p38MAPK signaling pathways.
