Seabuckthorn leaves are rich in phenolic substances,which have various effects such as anti-oxidation,antibacterial,hypoglycemic,and hypolipidemic effects.However,due to their poor water solubility,unstable and easy degradation,and unpleasant odor,its application are limited in the food field and as a waste resource it has not been fully exploited.Microencapsulation is an effective way to improve the stability,solubility and flavor of bioactive substances.In this study,based on the optimization of the extraction conditions of phenolic substances in seabuckthorn leaves,firstly,the extract of seabuckthorn leaves was encapsulated by electrohydrodynamics method.Based on the evaluation of structure and encapsulation effect,the focus was on the evaluation of the bioaccessibility and antioxidant activity of total phenols and flavonoids in seabuckthorn leaves extract and microcapsules during simulated digestion in vitro,as well as their effects on metabolic syndrome related enzymes(?-glucosidase,?-amylase and pancrelipase)to clarify the protective effect of encapsulation on bioactive substances.Secondly,the seabuckthorn leaf extract was applied to seabuckthorn oil and spray-dried to compare the similarities and differences of different additions of seabuckthorn leaf extracts(0.5 %,1 %,2 %)and the maximum amount of artificial synthetic antioxidants allowed by laws and regulations in protecting the oxidation stability of sea buckthorn oil;On this basis,2 % of seabuckthorn leaf extract was applied to the encapsulation of seabuckthorn oil by electrospinning to study its effect on improving the stability of oil and enhancing the antioxidant activity of microcapsules.The results showed that the optimum extraction conditions were solid-liquid ratio of1:8,extraction temperature of 60 ?,ethanol concentration of 70 %.Under these conditions,the polyphenol content of the extract was 386.1 mg GAE/g.SEM,FTIR and TGA showed that the extract was successfully encapsulated,and microencapsulation improved the tolerance of the active substance to the digestive environment(p H and digestive enzymes,etc.),thereby reducing its degradation rate and promoting the controlled release of the encapsulated compound and helping to increase their bioaccessibility in the intestine;In addition,compared with the free extract,the microencapsulated extract retained higher antioxidant activity and inhibitory effect on metabolic syndrome-related enzymes after digestion,which was beneficial to its hypoglycemic and hypolipidemic function in the human body.In the application of encapsulating sea buckthorn oil,sea buckthorn leaf extract could effectively improve its oxidative stability and enhance its antioxidant capacity,the effect was better than BHA and BHT and was positively related to the concentration.The results of this study showed that seabuckthorn leaf extract was feasible to be used as nutritional supplements and functional enhancers.On the basis of further study on its function,safety and application effect in different foods,seabuckthorn leaf was expected to play a role in the future food industry thereby avoiding waste of resources.
