Obesity has become an urgent global health problem,which is closely related to some metabolic diseases.Preventing obesity by adjusting dietary structure is one of the crucial ways to alleviate this health problem.In recent years,studies on the improvement of obesity and the regulation of intestinal flora by dark tea have emerged in an endless stream,which provides a theoretical basis for the application of dark tea in the intervention of obesity.Eurotium cristatum and Monascus can produce a variety of beneficial metabolites.Therefore,in this paper,raw dark tea is used as raw material,respectively inoculated with Eurotium cristatum and Monascus,and simultaneously inoculated with these two bacteria for liquid fermentation,and inoculated with Eurotium cristatum,Monascus,Eurotium cristatum and Monascus The tea soup was named ECF,MSF and COF,and the non-inoculated tea soup was named RF.The metabolites and volatile components of the fermented tea soup were analyzed and the tea soup was applied to the rat obesity intervention experiment induced by high-fat diet.(1)After the raw dark tea is fermented with Eurotium cristatum,Monascus,Eurotium cristatum and Monascus,its flavor will change because of the significant difference in volatile components between them.RF can only detect 14 main aroma components,which are mainly composed of some single esters,aldehydes and ketones,while the overall aroma components of ECF(36 types),MSF(33 types),and COF(36 types)become rich,which shows that the liquid fermented raw dark tea has a unique sensory experience.(2)To further evaluate the metabolic functional components in different fermented dark tea soups,it is found that the main components of the RF are amino acids and glycosides.Compared with RF,ECF has rich anti-obesity catechins metabolic active substances,MSF has rutin,caffeine and other antioxidant active substances,COF did not detect too many catechins,but the detection Flavonoids such as vitexin,luteolin,and daidzein have anti-inflammatory and antibacterial effects.(3)To further evaluate the obesity intervention function of dark tea and different fermented dark tea soup,60% high-fat diet was used to induce obesity in C57BL/6J mice.The high-fat diet group was named HFD,and the general diet group was named ND,After being fed with high-fat diet for 8 weeks,HFD body weight is 1.2 times that of ND and the difference was obvious(p<0.05)(Same below).After 7 weeks of intervention with tea soup,Compared with HFD,The weight loss of COF and MSF mice is obvious(p < 0.05).ECF weight loss is extremely obvious(p < 0.01).The results showed that the tea of RF,ECF,MSF and COF could interfere with the body weight of obese mice(4)The basic serum indicators were determined.ECF,MSF,and COF showed a certain regulatory effect on the serum lipid levels of obese mice.Compared with HFD,the levels of LDL-C,ALT,and AST decreased to varying degrees,while TC,TG showed a obvious decreasing trend,HDL-C obviously increased,among which ECF obviously Elevated the content of HDL-C compared with ND,and obviously reduced the content of AST compared with HFD.(5)The analysis of the mass of rat liver,epididymis and subcutaneous fat found that fermented tea soup can alleviate fat accumulation to varying degrees,and significantly reduce liver and fat mass compared with HFD.Among them,rat liver and epididymal fat after ECF intervention There is no obvious difference between subcutaneous fat mass and ND,and it has the best effect on improving fat accumulation in obese rat.(6)In addition,we also explored the intestinal health of obese rat,and found that the cecal flora of obese mice after the intervention of experimental tea soup has improved in varying degrees.The test tea soup improves the abundance of the dominant intestinal flora Firmicutes and Proteobacteria slightly.Compared with HFD,the test tea soup has a better intervention effect on liver lipid accumulation and obesity caused by diet.The abundance of Lactobacillus ? Alistipe flora;comprehensively,the experimental tea soup has a certain improvement effect on the abundance of beneficial microbial flora in the cecum,and MSF shows a better regulating effect.(7)Finally,in order to reveal the possible reasons for the effect of different tea soups on the intervention of animal obesity,we used Illumina Mi Seq sequencing to analyze the fungal community diversity at the stage of fermenting raw dark tea with mixed strains.The results show that during the COF mixed fermentation process,Monascus has always been dominant.This result explains that the cooperative COF mixed fermented tea soup is not as effective as ECF for obesity in rat.The possible reason is that Monascus,which has always been dominant,inhibits the growth of Eurotium cristatum and thus inhibits the fermentation metabolism of Eurotium cristatum.Produce anti-obesity active substances.
