Skin aging is a part of the overall aging of the body,mainly manifested in the generation of wrinkles,color spots,decreased skin elasticity,etc.With the increasing market demand of natural and multifunctional anti-aging products,the anti-aging activity of peptides and plant extracts has become a hot spot.At present,most studies focus on the evaluation of the activity of a single component.Therefore,the system has screened out food derived peptides and plant extracts which have better anti-aging effects,and made them into compound cream,to explore whether the two have synergistic effects,which is of great significance for developing safe and effective anti-aging active products.In this paper,three kinds of food-derived peptides(wheat peptide,soybean peptide,sea cucumber peptide)and nine kinds of food-derived plant extracts(rose eggplant powder,lotus leaf alkali powder,chrysanthemum powder,cassia seed powder,apigenin powder,rose powder,herbal tea powder,black tea powder,green tea powder)were selected as the research objects.By using in vitro tyrosinase inhibition test,bacteriostasis test and in vitro cell model test,active peptides and plant extracts with the best sunscreen and antioxidant function and they were made into compound cream.The skin aging function of compound cream was verified by mouse D-galactose induced aging model.The main results are as follows:(1)In vitro biochemical methods were used to screen peptides and plant extracts with antiaging related functions: UV scanning method and UV absorbance method were used to compare the sun protection function of peptides and plant extracts.It was found that only black tea powder and green tea powder had obvious absorption peak at about 270 nm,and 1 mg / ml black tea powder and green tea powder could achieve complete UV protection and highefficiency UV protection The results showed that DPPH free radical scavenging test,FRAP method and ABTS free radical scavenging test were used to compare the antioxidant function of the peptide and plant extract.The antioxidant function of green tea powder was the strongest,and the DPPH free radical scavenging rate IC50 was only(5.60 ± 0.48)? g / ml,The total antioxidant capacity measured by FRAP method was(8.69 ± 0.17)mmol / g,that measured by ABTS method was(1.91 ± 0.0046)mmol / g,followed by black tea powder.Among the three active peptides,sea cucumber peptide was the strongest,soybean peptide was the second,and wheat peptide was the worst.According to the above results,wheat peptide,soybean peptide,sea cucumber peptide,black tea powder and green tea powder were selected for further study.(2)Cell level analyzes and compares the anti-aging related functions of peptides and plant extracts: L929 fibroblast cycle test,apoptosis test and B16 melanoma cell proliferation inhibition test were used.It was found that when 200 ? g / ml sea cucumber peptide acted on L929 fibroblasts and B16 melanoma cells,the cell proliferation index and apoptosis rate of L929 fibroblasts were determined by The proliferation rate of B16 melanoma cells decreased to(72.7 ± 2.1)%.There was a significant difference between the blank control group and the blank control group(P < 0.05).Black tea powder and green tea powder had no significant effect on the cell cycle and apoptosis rate of L929(P > 0.05);compared with the blank control group,10 ? g / ml black tea powder and green tea powder could reduce the proliferation rate of B16 melanoma cells to(79.2 ± 0.94)% and(71.6 ± 1.0)% respectively.Therefore,the final screening results are sea cucumber peptide and green tea powder.(3)animal experiment is conductedt o evaluate the anti skin aging effect of Compound Cream: sea cucumber peptide cream,green tea powder cream and a compound cream mixed in equal proportion of them both are prepared and acted on D-galactose induced aging model mice'shaved backs for 7 weeks.The results showed that the catalase activity,superoxide dismutase activity and skin hydroxyproline content in the skin homogenate of the composite group were(10.1 + 0.50)U/mgprot,(189 + 15)U/mgprot,and(6.81 + 0.95)g/m L respectively,which were1.08 U/mgprot,33.9 U/ mgprot and 0.134 g/m L respectively,higher than that of the sea cucumber peptide cream and green tea powder cream.The above results show that the combination of sea cucumber peptide and green tea powder has synergistic effect,which is worthy of further study.
