Oryzanol is a mixture of sterol ferulate and cyclopineapple alcohol ferulate.Because of its extensive pharmacological effects and application value,it has great application value in medicine,food and chemical industries.At the same time,it It has also been a hot spot for scholars at home and abroad.Rice bran oil contains a relatively high content of oryzanol.During the saponification process,most oryzanol will be enriched in soapstock.The extraction of oryzanol from soapstock is also an important source of oryzanol.In this paper,rice bran oil soapstock was used as the raw material to extract and purify oryzanol through ethyl acetate enrichment extraction,methanol alkaline environment dissolution,acidic environment precipitation and other steps.The oryzanol extraction rate and purity were used as indicators to optimize the extraction process;Oryzanol as raw material,developed cream cosmetics containing oryzanol,and evaluated the sensory,physical and chemical indicators,skin feel,moisture retention and antioxidant capacity of this type of cosmetics,laying a foundation for the industrial production of oryzanol in the chemical industry.The main research results are as follows:1.Using ethyl acetate as the solvent,extract the oryzanol in the soapstock to obtain an ethyl acetate extract rich in oryzanol.The extract is concentrated by rotary evaporation to obtain an ethyl acetate concentrate;and the oryzanol is dissolved in Alkaline methanol is insoluble in acidic methanol.The ethyl acetate concentrate is purified,and then petroleum ether is washed to remove fat-soluble substances such as fatty acids and waxes.The oryzanol is purified by washing with warm water to remove salt.The extraction rate of oryzanol is used.And purity is the index,and the optimized process is as follows: ethyl acetate extraction times are3 times,alkaline methanol p H is 10,acidic methanol p H is 3,acidic methanol and petroleum ether washing times are 3 times,and the extraction rate of oryzanol is obtained.It is 52.63% and the purity is 94.03%.2.Based on the HPLC detection method,the best method and conditions for detecting the content of oryzanol in soapstock were established.The result is: the detection conditions of HPLC method are ethyl acetate as solvent extraction,chromatographic column is ZORBAX NH2 column,fluidity is ethanol,flow rate is0.6 ml/min,detection wavelength is 324 nm,under the above conditions,oryzanol is 4-50 μg The linear relationship within /ml is good,R2=0.9999,the relative standard deviation of this method is within 2%,and the average recovery rate is99.88%.This method for quantitative determination of oryzanol is simple,fast,sensitive and accurate.3.Add oryzanol to gel and cream cosmetics,and evaluate the sensory,physical and chemical indicators,skin feel,moisturizing properties and antioxidant capacity of the cosmetics.The results show that gels and creams The product has no oil-water separation phenomenon under cold and hot conditions,and has good stability;after use,the skin moisture content increases by 16.6% and 25.35%respectively.Gels and creams have good moisturizing properties.The gel and cream skin feel evaluation index scores are above 8 points,and have a good skin feel.When the concentration of oryzanol gel is 8 mg/ml,the DPPH free radical scavenging rate can reach 88.98%,and the gel has good antioxidant capacity.
