"Sudan Red" is not a food additive,but a chemical coloring agent commonly used by people.It contains a compound called naphthalene in its chemical composition,and naphthalene has an azo formula.The chemical structural properties of naphthalene make it a strong carcinogen,and it is highly toxic to the liver,kidneys and other human organs.Sudan red is mainly used as a color enhancer in industrial petroleum,motor oil and some other solvents.It is also used as a brightener for floors,shoes,etc.,also known as "Sudan".The dried chili in the hot pot base will fade for a long time.After adding Sudan Red,the hot pot base will remain bright red for a long time and will not fade for a long time.Once a person consumes foods containing Sudan Red,Sudan Red can cause cell cancer in the human body.If a large amount is taken in a short period of time,the most serious case will directly lead to death of the eater.Because the hot pot bottom material has a complex matrix and traditional methods are not efficient,it is of great significance to establish a fast and simple analytical method for the determination of Sudan red and apply it to practice.This paper is divided into two parts: The first part summarizes the formation mechanism,toxicity research and detection methods of Sudan Red.The second part is to determine the Sudan red contained in the hot pot bottom material by improving the pretreatment experimental method,and select the appropriate liquid chromatography conditions.A new type of solid phase extraction-high performance liquid chromatography method for the detection of Sudan red was explored.This chapter includes the selection of extraction solution and the activation of solid phase extraction cartridges.Established HPLC conditions for the determination of Sudan red in hot pot bottom materials,in which liquid chromatography column: Phenomenex Kinetex100×4.6mm,2.6?m,100A;column temperature: 40°C;mobile phase solution:(solvent A:acetonitrile: 0.1% formic acid Aqueous solution = 15:85.Solvent B: acetone: 0.1% formic acid in acetonitrile = 20:80.Solvent C: acetone);gradient elution,flow rate: 1.0 m L/min.Injection volume: 10?L.The liquid chromatographic detector is a diode array detector,and the detection is carried out after scanning the wavelength to obtain Sudan ???: 520 nm.Using this method,the linear relationship is better,the relative standard deviation(RSD)is between 1.20 and 1.74,and the average recovery is between 90.1% and 98.6%.The operation is more convenient than the national standard method.The four Sudan red peaks are separated cleanly and the peak detection time is three quarters shorter than the original standard.The detector has high sensitivity,and the analysis speed is more accurate and fast.In summary,Sudan red is toxic and seriously harmful to human health;the SPE cartridge pretreatment method and liquid phase detection method established in this study are simple and fast,with good reproducibility,low detection limit,and can quantitatively and qualitatively detect hot pot bottoms.Central Sudan Red.Compared with the GB/T 19681-2005 liquid chromatography method,this method is more convenient to operate and has higher recovery.
