Phyllanthus emblica is a commonly used Chinese herbal medicine for both medicine and food,which is widely cultivated in southern China.Phyllanthus emblica leaf is an additional product of Phyllanthus emblica fruit for both medicine and food,which is rich in resources.However,its modernization research is still relatively small,and it has not been effectively and widely used.Gallic acid is a kind of phenolic acid with a variety of pharmacological activities,which can be seen in the fields of food,medicine,chemical industry,electronics and so on.Gallic acid is the main phenolic acid active component in Phyllanthus emblica.The study on its extraction and separation can not only expand the utilization range of Phyllanthus emblica,but also increase the source of gallic acid extraction raw materials.In this paper,solvent reflux,acid hydrolysis and alkali hydrolysis were compared,and the acid hydrolysis method with the largest amount of gallic acid extraction from Phyllanthus emblica leaves was selected for further response surface optimization.Firstly,the solid-liquid ratio,hydrochloric acid concentration,extraction temperature and hydrolysis time were selected as single factors for the experiment.The results showed that the solid-liquid ratio of1:50 mg/m L,hydrochloric acid concentration of 4 mol/L,extraction temperature of 90?and extraction time of 1:50mg/m L were the best The results showed that the best extraction conditions were as follows:the ratio of material to liquid was 1:52.44mg/m L,the concentration of hydrochloric acid was 3.65mol/l,the extraction temperature was 100?,the extraction time was 3.31h,the predicted extraction amount was 34.11mg/g,the actual extraction amount was 33.91mg/g;Comparing the static adsorption and elution effects of NKA-9,xda-8,nka-2 and D301 resins,D301 resin was selected and used to further optimize the dynamic separation and purification conditions of gallic acid from Phyllanthus emblica leaves.The results showed that the separation and purification conditions were as follows:loading concentration 0.23mg/m L,loading speed 2BV/h,loading volume 13BV,eluent 70%ethanol,elution speed 2BV/h Under the optimal conditions of adsorption and elution,0.149g lyophilized sample was obtained.The yield of the final product was 13.18%,and the content of gallic acid was 27.63%;The thermodynamics and kinetics of gallic acid adsorption by D301 resin were studied under static adsorption conditions.It was found that the adsorption thermodynamics and kinetics of gallic acid by D301 resin were better fitted with Freundlich equation and quasi second order kinetic model,and the equations were:lg Q_e=0.748lg C_e+2.3672 and T/Qt=0.1143t+2.0214,respectively;Finally,an antibacterial and anti-inflammatory mouthwash containing gallic acid extract of Phyllanthus emblica leaves was prepared.The technological conditions were as follows:the amount of extract was 0.20%,the amount of glycerol was 10%,the amount of ethanol was 1.5%,the amount of xylitol was0.2%,and the amount of menthol was 0.015%.The inhibition rate of oral bacteria between the commercial mouthwash and the self-made mouthwash was compared.Results the average inhibition rate of self-made mouthwash was 17.62%,The average antibacterial rate of commercial mouthwash was 14.88%,and the result of self-made mouthwash was slightly better.
