In recent years,fluorescent probes evolved from organic fluorescent materials have attracted wide attention because of their excellent selectivity,high sensitivity,and rapid response.and their unique fluorescence properties have shown important research value in the fields of bioimaging,biomolecular recognition and diagnosis.However,in the biological field,the development of traditional fluorescent probes is limited due to their poor water solubility,complex molecular design,poor storkes shift,aggregation fluorescence quenching,and poor cell membrane penetration,etc.Therefore,this paper further designs,improves and explores the shortcomings of the above traditional probes.The main research are listed as following:(1)Preparation and application of CyssTPN fluorescent probe based on integration of tumor diagnosis and treatmentTriptolide(TP)has been shown to be an effective anti-cancer drug to treat a wide range of cancers.However,it is limited by the high toxicity,poor solubility and poor bioavailability of triptolide.In order to overcome the disadvantages of TP,the hydrophilic Cy(IR780)and hydrophobic TP were synthesized by disulfide bond to form organic small molecules(Cyss TP).Then,Cyss TP self-assembly to prepare the organic fluorescent probe(CyssTPN)in the aqueous phase.Through transmission electron microscope(TEM)characterization,it had proved that the average size was 50 nm.According to cell uptake,cytotoxicity and simulated tumor therapy,CyssTPN had the ability to track the release of TP in real time,and applied for tumor therapy effectively.The results showed that CyssTPN fluorescent probe could improve the solubility of TP,but also reduce its side effects and improve the bioavailability of TP.As a simple molecular structure,the fluorescent probe had enhanced fluorescence intensity and tumor targeting,which improved the effect of integration of tumor diagnosis.(2)Preparation and bioimaging application of water solubility aggregation induced fluorescent nano-probesThis paper modified(5-formylfuran-2-yl)boric acid?4-(1,2,2-triphenylvinyl)benzoic acid?3,4-dimethoxyphenylboric acid and 4-aminophenylboric acid by Suzuki coupling reaction to modify 4,4'-(4-bromobenzene)methylene)bis(N,N-dimethylaniline),Then,oxidation reaction to form a series of organic fluorescent probes(MG-A?MG-B?MG-C and MG-D).Mg-A was selected for analysis and verification.The results showed that it had good water solubility and fluorescence properties in the form of aggregation state when the volume ratio H2O/THF(1:10),which showed AIE characteristics.The DFT/B3LYP/6-31G(d)calculation of Gaussian 09 further proved that the change of fluorescence was generated by ICT mechanism.The spherical particles with an average diameter about 20 nm were obtained according to TEM and other characterization.By pH testing and cell imaging,it showed that it had pH response and mitochondrial targeting,which reflected its good biocompatibility.
