Objective This project intends to develop a daily cosmetics based on the extract of Phellinus igniarius,which is a kind of medicinal and edible fungus.Considering that Phellinus igniarius has not yet entered the catalog of cosmetic raw materials,the safety evaluation of Phellinus igniarius as a cosmetic raw material was conducted in accordance with the"Cosmetic Hygiene Regulations"(2015 Edition)and"Food Safety Standards";and the cosmetics with Phellinus igniarius extract added through animal experiments Perform efficacy verification and quality evaluation to evaluate the safety,effectiveness and usability of Phellinus linteus in cosmetics.Method1.Preparation and activity screening of Phellinus igniarius samples:using Phellinus igniarius strains of Phellinus igniarius fermentation broth and Phellinus igniarius fruiting bodies as raw materials,the fruiting bodies are extracted by ethanol,water and different organic solvents to obtain extracts.And extracts,the bacterial liquids of different fermentation days are extracted with ethyl acetate to obtain the bacterial liquid extracts of different fermentation days,the total antioxidant capacity of the fruit body and the bacterial liquid samples,the inhibition rate of tyrosinase activity,the absorbance in the ultraviolet band,etc.The indicators are measured,the most suitable sample as a cosmetic raw material is screened,and the free radical scavenging rate of the sample is tested,and the UV absorption capacity and total polyphenol content are analyzed.2.Preliminary evaluation of the safety of Phellinus igniarius and Phellinus igniarius extracts:animal experiments were used to conduct acute oral toxicity and acute dermal toxicity tests on the extracts;the raw material Phellinus igniarius fruit bodies were tested for 90-day oral toxicity.3.Development of Phellinus igniarius cosmetics and preliminary evaluation of their quality and efficacy:Optimizing the ratio of the water and oil phases of the cosmetics through orthogonal experiments and sensory evaluation,and designing Phellinus igniarius day cream with a good sense of use;microbiological testing and heavy metal testing for the day cream The limit and other indicators are measured,and the anti-ultraviolet radiation and anti-aging effects of the day cream are evaluated through animal experiments.Result 1.Preliminarily obtained 10 samples of Phellinus igniarius to be tested,and comprehensively evaluated the total antioxidant capacity of each sample,the inhibition rate of tyrosinase activity,and the absorbance of ultraviolet band to screen out the ethanol extract of Phellinus igniarius(SH2)as cosmetic raw materials.The scavenging rate of SH2 on free radical DPPH is as high as89.7 0.00%;when the concentration is 125?g/mL,the absorbance of SH2 in the UVB band is higher than 2.0;the total polyphenol content accounts for 60.4%.2.The acute oral toxicity and acute dermal toxicity test of SH2,and the 90-day oral toxicity test of Phellinus igniarius fruit body,all experimental indicators are within the normal range,indicating that SH2 and Phellinus igniarius are actually safe,non-toxic and non-irritating.3.The main raw material ratio of the water phase of the oil-in-water Phellinus igniarius day cream is 5%glycerol,8%butanediol,2%propylene glycol,and 2%sorbitol;the main raw material ratio of the oil phase is 1%lanolin,4%petroleum jelly,Huohuo Brazil oil 5%,glyceryl stearate 1%.The produced Phellinus igniarius day cream has good stability,and the microbiological testing and heavy metal limit testing meet the standards.In the evaluation of efficacy,the enzyme index detection in the aging skin showed that the content of SOD and CAT in the day cream group was significantly different from that of the model group(P<0.01),and the content of HYP and MDA was significantly different(P<0.05).Ultraviolet radiation skin damage score showed that the damage degree of the day cream group was significantly different from that of the model group(P<0.01).In summary,Phellinus igniarius is safe and non-toxic,and the quality of Phellinus igniarius day cream is stable and has certain UV protection and anti-aging effects.
