Resistant starch(RS)can not be digested and absorbed in the small intestine of healthy people,but can only be metabolized by microorganisms in the large intestine.Its degradation products have physiological functions such as hypoglycemia,Hypolipidemia,weight control and prevention of intestinal diseases.Resistant starch also has the function of dietary fiber.Because of its good processing characteristics,it has broad development prospects and has gradually become a research hotspot in recent years.In this paper,Mung bean resistant starch(MRS)was prepared from mung bean starch by microwave enzymatic method.Taking the content of resistant starch as the index,on the basis of single factor experiment,the optimum preparation conditions were determined by response surface methodology.The resistant starch prepared by this method was fed to mice,and its effects on intestinal metabolism were studied.In vitro test is a supplement to animal test to study the effect of MRS on intestinal flora.In order to produce a functional staple food which can control body weight,improve intestinal balance,increase satiety and reduce intake,the resistant starch was added to high gluten wheat flour for bread making.The main conclusions are as follows:(1)Preparation of mung bean resistant starchThe optimum conditions of microwave enzymatic method were as follows: starch concentration 24%,microwave power 750 W,microwave time 110 s,?-amylase dosage 610 U/g,?-amylase reaction time 54 min,pullulanase dosage 25 U/g,pullulanase reaction time 7 h.Under these conditions,the highest content of resistant starch was 35.83%.(2)Effects of mung bean resistant starch on intestinal metabolism and blood lipid in mice(1)MRS can control the weight gain of mice,increase satiety,reduce food intake and reduce feed utilization.(2)MRS can effectively reduce the p H value of cecum and increase the water content of its contents.(3)MRS can increase the content of acetic acid,propionic acid,butyric acid,isobutyric acid and other short chain fatty acids,especially in the high-dose group.(4)MRS can effectively reduce the content of total cholesterol(TC),total triglyceride(TG),high density lipoprotein cholesterol(HDL-C)and low density lipoprotein cholesterol(LDL-C)in serum.The decrease of arteriosclerosis index(AI)can prevent arteriosclerosis.(3)Formula optimization of mung bean resistant starch breadTaking sensory score as index,single factor experiment was used to study the effects of mung bean resistant starch addition,yeast dosage,sugar dosage and corn oil dosage on bread quality.The formula was optimized by orthogonal test.The optimum conditions were as follows:MRS addition 9%,yeast dosage 2.5%,sugar dosage 4%,corn oil dosage 8%,Under these conditions,the highest sensory score was 89.2.
