Antarctic krill(Euphausia superba)is an important biological resource in the ocean.The protein,oil,and polysaccharides in Antarctic krill are widely used in food,medicine and chemical industries.Antarctic krill oil has a complex combination of multiple active ingredients and has synergistic biological activity.The content of polyunsaturated fatty acids is as high as 40%.The main ingredients are(EPA,C20: 5)and(DHA,C22: 6),and EPA and DHA It exists in the form of phospholipids,has higher bioavailability,has many functions such as anti-inflammatory,preventing obesity,hyperglycemia,enhancing immunity and memory.This study explored the potential efficacy of Antarctic krill oil from the extraction and preparation of Antarctic krill oil,cell inflammation experiments and animal model experiments,and provided a theoretical reference for the research and development of Antarctic krill oil.Research indicates:(1)Antarctic krill are pretreated to obtain dried shrimp powder,freeze-dried shrimp powder and frozen shrimp.Antarctic krill oil is prepared by organic solvent extraction and supercritical carbon dioxide extraction,and chloroform methanol in organic solvent extraction The yield of oil is the highest,followed by ethanol and acetone,and n-hexane is the lowest.The yield of krill oil obtained by supercritical carbon dioxide extraction method is lower than that of organic solvent extraction method.However,its physical and chemical indicators are better than that of Antarctic krill oil extracted by organic solvent.Supercritical freeze-dried shrimp oil detected 29 fatty acids,with the most types.Its saturated fatty acid content was25.04%,monounsaturated fatty acid content was 29.65%,polyunsaturated fatty acid content was 42.40%,and astaxanthin was as high as 589?g/g.(2)Using LPS to stimulate the Raw264.7 cell inflammation model,the in vitro anti-inflammatory activity of supercritical freeze-dried shrimp oil and refined freeze-dried shrimp oil was studied.The results showed that both can inhibit LPS-induced promotion at the transcription and translation levels.The transcription and expression of inflammatory cytokines IL-1?,IL-6 and TNF-?.Western Blot experiments have proved that they can inhibit the phosphorylation of I?B? protein,inhibit the phosphorylation of NF-?B protein,and inhibit the NF-?B signaling pathway.Activate,and ultimately inhibit the production of pro-inflammatory factors IL-1?,IL-6 and TNF-?,and play an anti-inflammatory effect;supercritical freeze-dried oil has a better anti-inflammatory effect than refined freeze-dried shrimp oil.(3)Through the establishment of ethanol-induced acute gastritis model in rats,the effect of krill oil on gastric mucosal damage was explored.The results showed that krill oil can reduce oxidative stress and lipid peroxidation,and inhibit the phosphorylation and degradation of I?B?.The transcriptional activity of NF-?B reduces the production of inflammatory factors and reduces the inflammatory response of gastric tissue.
