In recent years,the occurrence of fatty liver disease in China is increasing day by day,which not only affects the normal life of patients,but also brings great burden to the family economy of patients.At present,the treatment of fatty liver disease mainly adopts adjustment of living habits while taking certain dietary intervention or liver protection drug intervention.Studies have shown that fructus embalsam,pitaya fruit,sea buckthorn and other fruits have A certain effect on liver protection.Therefore,this study used two kinds of beverages fermented from these fruits(fermentation liquid A and fermentation liquid B)to explore their effects on alcoholic fatty liver and metabolic fatty liver and their effects on gut microflora.In this study,Kunming mice were used as experimental subjects,alcoholic liver disease(ALD)model was established by daily ethanol intragastric administration while alcohol was added to drinking water,metabolic fatty liver(MAFLD)model was established by high-fat diet feeding,and different fermentation broths were given intragastric administration according to groups every day.After modeling,serum aspartate aminotransferase(AST),alanine aminotransferase(ALT)and total bilirubin were detected to evaluate the degree of liver injury.The degree of liver fibrosis was evaluated by detecting four hepatic fibrosis parameters and masson staining results of liver slices.The antioxidant capacity of liver was detected.The expression of triglyceride,total cholesterol and GP73 protein in liver were detected,and liver lipid accumulation and pathological changes were evaluated by H?E staining.In addition,the activities of liver alcohol dehydrogenase(ADH)and alcohol dehydrogenase(ALDH)were detected in ALD group to evaluate the ability of liver to cure alcohol.At the same time,gut microflora 16 S r DNA metagenomic analysis was performed on cecal contents of each group of mice to explore the mechanism of the influence of two kinds of fermentation broth on fatty liver development through gut microflora.The results showed that fermentation liquid A can improve the function of anti-hangover effects or activity,reduce the influence of alcohol on body weight and liver weight,and reduce the degree of liver injury.It has A good intervention effect on liver fibrosis and cirrhosis caused by long-term alcohol intake,and can even delay the occurrence of early liver fibrosis.B fermentation broth can improve liver lipid metabolism,reduce the influence of high fat diet on body weight and liver weight,and reduce the degree of liver injury;Both fermentation liquid A and B can effectively alleviate the problem of weakened antioxidant capacity of liver caused by alcohol and fat accumulation,alleviate the damage of liver caused by long-term alcohol intake and high-fat diet,and have certain effect on inhibiting hepatocellular lesions of ALD and MAFLD.gut microflora analysis,according to the results of A and B can improve ALD and MAFLD mice gut microflora species diversity,and for some harmful bacteria such as Erysipelotrichales,Coriobacteriales,Desulfovibrionaceae,Deferribacteracea and other inhibition,at the same time promote some beneficial bacteria such as Lactobacillus,Ruminococcaceae,Lachnospiraceae,Lactococcus,Akkermansia,to improve gut microflora disorder caused by MAFLD.The two fermentation broths interfered with the occurrence and development of ALD by regulating the bacteria related to ethanol metabolism,fatty acid synthesis and the tricarboxylic acid cycle.The development of MAFLD can be interfered by regulating NAD rescue and glycogen metabolism.Conclusion:1.Both fermentation liquid A and B can effectively intervene in the development of ALD induced by alcohol in mice,and the effect of fermentation liquid A is better than that of fermentation liquid B.2.Both fermentation liquid A and B can effectively intervene in the development of MAFLD induced by high-fat diet in mice,and the effect of fermentation liquid B is better than that of fermentation liquid A.3,A,B two kinds of fermented liquid can increase the species diversity of gut microflora in mice,inhibit such as erysipelas bacteria,red bugs,bacteria,vibrio for desulphurization,iron bacteria and other harmful bacteria growth,promote the lactobacillus,milk coccus,Mr Ackermann bacteria,rumen bacteria,MAO spirillum beneficial bacteria breeding,such as health,regulating the gut microflora intervention ALD and MAFLD development.
