Whitening is the act of brightening your skin visually by using makeup to modify the natural color of your skin,or by long-term use of products that contain less melanin in your skin.Whether the added whitening ingredients can effectively reduce the production of melanin is a very important link in the research and development of whitening cosmetics.The study of whitening is not only for the visual beauty of skin,but also for the prevention and treatment of skin diseases.Consumers for whitening cosmetics have high enthusiasm for consumption,in order to better ensure the rights and interests of consumers,standardized regulation is particularly important.The evaluation results from the same model are more referential and comparable,and are helpful for enterprises in the industry to conduct self-examination,as well as the proven effectiveness of cosmetics can also be used as a benchmark for the industry to spur common progress of the entire market.The main research contents and results of this project are as follows:1.In this study,a model experiment method of mushroom tyrosinase was designed for high-throughput research,the activity and efficacy of mushroom tyrosinase were evaluated by?-arbutin(ARB),raspberry ketone(RK),raspberry ketone glucoside(RKG),3-O-Ethyl Ascorbic Acid(EA),Glabridin(GLA),Glycyrrhetinic Acid(GA)and Dipotassium Glycyrrhizinate(DG),all of them had inhibitory effect on mushroom tyrosinase.Except GA and DG,all of them had inhibitory effect on mushroom tyrosinase.Among them,the inhibition efficiency of ARB,EA and GLA at the highest concentration were all higher than 50.00 %.RK was similar to RKG and the inhibition rate was close to 30.00 %.2.In this study,we designed a B16 mouse melanoma cell model for high-throughput study by referring to the literature,the melanin content and the inhibitory effect of tyrosinase activity were measured by ARB,RK,RKG,EA,GLA,GA and DG.Among the 7 effective ingredients of whitening,the other ingredients except GA had melanin inhibiting effect.Ga at the highest concentration can promote the formation of cell melanin.Among the seven active ingredients of whitening,ARB,GLA,EA,GA and DG all had the inhibitory effect on cell tyrosinase,and the inhibitory rate of DG was the highest(88.64 %).RK,RKG and EA did not inhibit tyrosinase activity.3.In this study,an experimental method for evaluating the whitening efficacy of highthroughput zebrafish embryo model was designed by reference,the melanin content and tyrosinase activity of zebrafish were measured by ARB,RK,RKG,EA,GLA,GA and DG.There was no effect of 7 kinds of whitening ingredients on the development of zebrafish embryo,and all of them developed normally.Among 7 kinds of whitening active ingredients,ARB,RK,RKG,GA all had the inhibitory effect on the melanin content of zebrafish model.The inhibition efficiency of ARB,RK and RKG at the highest concentration were all higher than 50.00 %.GA has a weak inhibition effect.The inhibition rate is only 28.71 %.The inhibitory effect of ARB,RK,RKG and GA on tyrosinase activity in zebrafish model.The inhibition efficiency of ARB,RK and RKG at the highest concentration were all higher than50.00 %.GA has a weak inhibition effect,the inhibition rate is only 31.60 %.4.In this study,a formula of emulsion matrix was designed.The amount of active ingredients in 6 groups were 3.00 % ARB,1.00 % RK,3.00 % EA,1.00 % GLA,1.00 % DG and 3.00 % RKG respectively.All the 7 emulsions have been tested and evaluated.The properties are stable and meet the basic requirements of the emulsions.The results showed that all the 7 emulsions were safe,and only the one with ARB added in the 6 emulsions had better effect of brightening skin color,which had whitening effect in human skin model.In addition to ARB,some of the other active ingredients have been used in whitening cosmetics,but in this trial did not show whitening effect.5.In this study,four whitening efficacy evaluation models were compared.According to the experimental results,the use of mushroom tyrosinase model alone may result in the false negative result of screening antipigmented components.The B16 Mouse Melanoma Cell Model had the highest positive rate.However,the production of melanin in B16 cells is very unstable,resulting in poor reproducibility of the results.This model is not an economic evaluation model for whitening efficacy because of the cost of repeated experiments.Zebrafish model,which can simulate the different stages of melanin maturation.By comprehensive comparison,zebrafish is the most suitable model for the pre-screening of active ingredients in whitening.Human efficacy model is the closest model to the real user experience,these 6whitening emulsions have no adverse reactions,but only add ARB whitening emulsions have the effect of brightening the skin color of the test site.The evaluation data of human efficacy model in this study have high reference value for subsequent commercialization of various whitening efficacy components.
