Agarwood is fragrant resinous heartwood from wounded tree of the Aquilaria and Grinops species.In order to enrich the skeleton structure of the compounds in agarwood,provide scientific basis for the further development and theoretical basis for the identification agarwood,the phytochemical investigation on agarwood originated from Aquilaria species was carried out.Twenty-five compounds were isolated by various columne chromatographic from the ethanol extract of agarwood.Their structures were identified by spectroscopic methods as follows:(+)-12,13-dihydroxyguaiol(1),rel-(2R,5R,10S)-6(7)-spirovetiven-11,12,13-triol(2),4-methoxy-11,12-hydroxy-5(6)-ene-zizaane(3),6-hydroxy-8-chloro-2-[2-(4’-hydroxyphenyl)ethyl]chromone(4),6-hydroxy-7-methoxy-2-[2-(3’-hydroxyphenyl)ethyl]chromone(5),aquilarias A-G(6-12),aquilasesquits A-E(13-17),6,7-hydroxyl-2-(2-phenylethyl)chromone(18),6,7-hydroxy-2-[2-(4’-methoxyphenyl)ethyl]chromone(19),6-hydroxy-7-methoxy-2-[2-(4’-hydroxy phenyl)ethyl]chromone(20),6-hydroxy-2-[2-(3’-methoxy-4’-hydroxyphenyl)ethyl]chromone(21),5a,6a,7/3,8a-tetrahydroxy-2-(2-phenylethyl)-5,6,7,8-tetrahydro-chromone(22),agarptetrol(23),5a,6β,7β,8α-tetrahydroxy-2-[2-(4’-methoxyphenyl)ethyl]-5,6,7,8-tetrahydrochromone(24),6-hydroxy-2-[2-(3’-methoxy-4’-hydroxyphenyl)ethenyl]chromone(25),including three sesquiterpenoids,ten 2-(2-phenylethyl)chromones,twelve bi-phenylethylchromones.Among them,seventeen new compounds,including three new sesquiterpenes(1-3),two new 2-(2-phenylethyl)chromones(4 and 5),and twelve were new bi-phenylethylchromones(6-17).The cytotoxic test of compounds 18-25 showed that compound 25 exhibited significant activity against K562 and BEL-7402 with IC50 values 9.25 and 15.32μM,respectively.While,compound 21 was exhibited weak inhibition against five human cancer cell lines,and compounds 18-20 and 22-24 were exhibited no activities against five human cancer cell lines.
