Agarwood is the resinous heart portion of some Aquilaria trees. It possesses variousbiological activities such as sedative, analgesic and digestive. The agarwood can not befound in healthy A.sinensis, but rather in wounded parts. But the mechanism of formationof agarwood is not clear. There are two parts of main chemical compounds in agarwood,volatility and non-volatility. Sesquiterpenes are the main constituents of volatile oil. The2-(2-phenylethyl) chromone derivatives are the other major constituents of agarwood. It isnecessary to establish methods to analyse the compounds. This article aims to establish theextracted and detected methods to know the main constituents in different samples. Thiswork plays an important role in learning the mechanism of agarwood formation andproducing high quanlity agarwood.Firstly the optimal method to extract the2-(2-phenylethyl) chromones was chosen. Atthe same time, the method to simultaneously determine four2-(2-phenylethyl) chromonesin agarwood by high performance liquid chromatography (HPLC) was established. Thesamples induced by artificial liquid transfusion technology were analyzed by HPLC. Itwill have important significance to the establishment of the evaluation method and therelated research.Then identified the2-(2-phenylethyl) chromones by liquid chromatograph massspectrometer (LC-MS). The ultra performance liquid chromatographyquadrupole-time-of-flight mass spectrometry (UPLC/Q-TOFMS) was used to identify2-(2-phenylethyl) chromones in different agarwood. Positive model was adopted. MS andMS/MS were conducted to provide abundant information for structure identifications and6compounds were found. The fast, accurate and effective LC-MS method was establishedto identity the2-(2-phenylethyl) chromones.The detection of the sesquiterpenes was mainly by gas chromatograph massspectrometer(GC-MS). Firstly compositions and antifungal activities of essential oils fromdifferent samples were investigated. The compositions of agarwood induced byLasiodiplodia theobromae and wild agarwood essential oils were both rich in sesquiterpenes. Essential oils of the samples were potent inhibitors of fungus. Then a fastextraction method, solid phase microextraction (SPME), was established to analyse theessential compounds in agarwood. The conditions of SPME method were optimized, and asample was tested and analysed by GC-MS and retention indices (RI). Finally, themechanism of agarwood formation in A.sinensis induced by Lasiodiplodia theobromaewas studied by SPME-GC/MS.
