Rapid and effective hemostasis has always been a challenge in the treatment of trauma.Natural halloysite nanotubes(HNTs),which has hollow tube-like nanostructure,is a kind of inorganic aluminosilicate material which has attracted much attention in recent years.In this study,RG5 peptides with hemostatic activity were loaded into halloysite nanotubes to prepare a new type of composite nano-hemostatic powder(HNTs/RG5).The micro-morphology of the hemostatic powder was characterized by transmission electron microscope(TEM).The hemostatic performance of the composite hemostatic agent in vitro was evaluated by TEG(Thromboelastogram)and coagulation test in vitro.The results showed that RG5 was successfully loaded into the lumen of halloysite nanotubes,and the clotting time of the same amount of nano-hemostatic powder was 268 seconds shorter than that of Celox?.Then the injectable gel dispersion system was prepared by blending HNTs/RG5 composite nano-hemostatic powder with sodium alginate / gelatin to form AGHR composite hemostatic hydrogel.The safety of AGHR compound hemostatic hydrogel was evaluated by skin irritation test,CCK-8 cytotoxicity test and blood compatibility test.The results showed that AGHR composite hydrogel had no irritation to animal skin,while had low cytotoxicity to L929 cells and good blood compatibility.The microstructure and rheological properties of AGHR hydrogel were analyzed by scanning electron microscope(SEM),viscometer,Fourier transform infrared spectroscopy(FTIR)and rheometer,and the hemostatic properties of AGHR composite hydrogel in vivo were evaluated by rat liver injury model.The experimental results showed that the AGHR composite hydrogel could maintain stable rheological properties in the range of 5-80 ℃.On the one hand,its layered and pore structure was beneficial to quickly stop bleeding after sealing or closing the wound,on the other hand,it was helpful to transport nutrients secreted by activated platelets such as cell growth factor to the wound surface in time and promote wound healing.The results of model animal experiments showed that AGHR composite hydrogel had better hemostatic effect compared with the blank control group,and the hemostatic time was shortened by 61%.We can conclude from this study that AGHR composite hydrogel is a relatively safe and superior injectable rapid hemostatic material,which is expected to play an important role in first aid and clinical rescue in the future.
