| Aronia melanocarpa fruit rich in polyphenols,flavonoids and anthocyanins it has health care functions such as anti-inflammatory,anti-tumor,anti-radiation,lowering blood sugar,etc.Used in food and drug development,So the choice of extraction method is very important.Currently Aronia melanocarpa extract mainly based on ultrasonic assisted extraction method,however,there are disadvantages such as long extraction time and temperature limitation.The flash extraction method is time-saving,high-efficiency,and is not limited by temperature.So this test is based on Aronia melanocarpa the fruit is the material,the flash extraction method isusedtoex tract polyphenols and flavonoids of Aronia melanocarpa,Use response surface method to optimize on the basis of single factor,determine the best extraction conditions and determine their antioxidant activity,the results are as follows:1.The best extraction conditions for flash extraction technology are:Extraction time 59.14 s,ethanol concentration 61.6%,solid-liquid ratio1:42.63 g·ml-1,under these conditions,the extraction rate of polyphenols is20.10 mg·g-1,the content of flavonoids is 2.96 mg·g-1.The optimal conditions for ultrasonic-assisted extraction are:extraction time 40 min,ethanol concentration 57.74%,extraction temperature 35.99℃,solid-liquid ratio1:25.6 g·ml-1,under these conditions,the polyphenol content is 19.41 mg·g-1.The flavonoid content is 3.39 mg·g-1.2.By measuring the free radical scavenging rate of DPPH,ABTS+and superoxide anion(O2-)on the polyphenols of Sorbus melanocarpa,evaluate the antioxidant activity of the polyphenols,and remove DPPH and superoxide.Anion(O2-)IC50value is higher than Vc IC50and the difference is significant,and the difference between the scavenging ABTS+free radical IC50value and Vc IC50is not significant. |
