In the 1990 s,the Aronia melanocarpa began to be widely planted in Liaoning,Jilin and other provinces.Its mature fruit is rich in anthocyanins,fats,proteins,carbohydrates and other major nutrients,and the anthocyanin content is about 25% of phenolic compounds.Anthocyanins have anti-oxidation,hypoglycemic,cardiovascular and cerebrovascular diseases,anti-tumor effects,etc.,and are widely used in health care products.However,anthocyanins are unstable to external factors such as light and heat,microcapsule technology is used in this paper to control the release of anthocyanins,prolong the storage period of anthocyanins,and enhance the stability of anthocyanin.The anthocyanin from the Aronia melanocarpa was extracted by ethanol extraction method,the extraction temperature,extraction time and material liquid ratio were optimized by response surface methodology to determine the optimal extraction.The best macroporous resin was selected from five macroporous resins of X5,D101,AB-8,HP-20 and XAD-7 by static adsorption for purification,we researched the following factors:sample concentration,ethanol tvolume fraction of the eluent,adsorption flow rate and desorption flow rate.The microcapsules of the anthocyanin from the Aronia melanocarpa were prepared by spray drying method.The core wall mass ratio,inlet temperature and feed amount were optimized by orthogonal test to determine the optimal microencapsulation process.On this basis,the stability and surface structure of the anthocyanin microcapsules from the Aronia melanocarpa were studied.The main research contents and conclusions are as follows:1.Anthocyanin extraction from Aronia melanocarpaAnthocyanins were extracted from the Aronia melanocarpa by ethanol extraction,By response surface optimization method,the optimal extraction conditions were obtained: ethanol volume fraction 60%(pH 2-3),extraction temperature 46?,material liquid ratio 1:23.8(g/mL),extraction time 4.2 h,the content of anthocyanin was 6.43(mg/g)by HPLC.2.Purification of anthocyanins from Aronia melanocarpa(1)Five macroporous adsorption resins X5,D101,AB-8,HP-20,XAD-7 were used to separate and purify the anthocyanins from the Aronia melanocarpa,the XAD-7 macropores resin is the best by static adsorption test,the adsorption amount is 1.67 mg/g,and the desorption rate is 87.96%.(2)Based on static adsorption test,the sample concentration is 0.45 mg/mL,and the 30% ethanolacid solution is the best.(3)In the dynamic adsorption test,the adsorption flow rate was 1.0 mL/min and the desorption flow rate was 1.0 mL/min.3.Aronia melanocarpa anthocyanin microcapsulesMicroencapsulation was conducted by spray drying,and the embedding ratewas obtained HPLC method.Orthogonal test was carried out to obtain the best process conditions: acacia/maltodextrin 2:3(w/w)for wall material,wall material to core material ratio of 2:8(v/w),inlet temperature of 120?,feed amount of 15 mL / min,its embedding rate is 92.89%.4.Characterization and stability study of anthocyanin microencapsulation in Aronia melanocarpa(1)The microcapsules were characterized by scanning electron microscopy.The surface of the microcapsules was smooth and spherical,and some of them were in the shape of folds or depressions.The surface had no cracks and the microcapsules had better effects.(2)By comparing the stability of light and heat of anthocyanins from the Aronia melanocarpa before and after microencapsulation,the results showed that the anthocyanin microcapsules enhanced the stability of light and heat,slowed release and prolonged storage time of the anthocyanins,the purpose of this experiment was achieved.
