| The proanthocyanidins contained in grape seed has a variety of biological functions,which are constituted as monomers and dimers(OPC,20%),poly procyanidins(PPC,80%),and the high polymer is hard to absorb,and the bioavailability is low.At present,it is currently a liquid acid catalytic method,and there is no continuous operation of equipment corrosion in the process of industrial production.This paper uses four factors(temperature,material-ratios,catalyst dosage,time)five level central combination response surface method to optimize the reaction parameters,and the screened zeolite Y is modified.The main findings are as follows:1.UPLC qualitative and quantitative analysis of grape seed proanthocyanidins extract(GSPE):G(1.6 min,1.0%),EC(4.0 min,2.0%),C(3.3 min,4.0%),ECG(8.1 min,0.7%),B1(2.6 min,1.4%),B2(3.0 min,1.1%),B3(3.0 min,0.4%),PPC(9.6 min,86.8%);hydrochloric acid-vanilla aldehyde method measuring GSPE:ethyl acetate extraction phase(the degree of polymerization(mDP)of 2.1,DPPH IC50 of 3.6 μg/mL),residua-laqueous phase(mDP of 5.8,DPPH IC50 of 4.6 μg/mL).2.Screening the mesoporous zeolite Y as a solid acid catalyst according to the PPC depolymerization rate and the condition by single factor experiment getting the depolymerization of GSPE by zeolite Y is:temperature of 130℃,mesoporous zeolite Y/GSPE ratio of 5,time of 18min,and the mDP is 3.5,the dgree of depolymerization is 30.3%,the concentration of B2 increases by 0.1 mg/ml;when the reaction system is added to EC(EC/GSPE=2),the mDP is 1.9,the degree of depolymerization is 35.5%,and the concentration of B2 increases by 1.9 mg/ml.3.The best condition of EC/mesoporous zeolite Y depolymerising GSPE is:temperature of 121.9℃,EC/GSPE ratio of 1.2,mesoporous Zeolite Y/GSPE of 4.4,time of 18.7min,the response value of B2 is 69.1 mg/g,the experimental value is 67.8 mg/g,the deviation is 1.4%;the best condition of EC-C/mesoporous zeolite Y depolymerising GSPE is:temperature of 130℃ EC/GSPE of 1.3,mesoporous zeolite Y/GSPE of 2.5,time of 13.5 min,B2 response value is 55.6 mg/g,experimental value is 54.6 mg/g,deviation is 1.8%.4.Using seven components to perform load modification finds that the PPC polymerization rate is up to 68.7%and the mDP is 1.7 when using 50%K2CO3/mesoporous Zeolite Y(mass ratio)of 500℃ for 4 hours. |
