Metal complexes containing nitrogen and oxygen heterocyclic organic ligands often show better DNA bonding or nuclease activity.This article uses 2,2’-bipyridine(bipy),1,4-terephthalic acid(H2BDC),1,10-phenanthroline(phen,),monoketone(DAFO),and pyromellitic acid as Ligand,copper complexes 1-4 were synthesized with transition metal copper,named:[Cu(DAFO)2]·(NO32]·2NH3·H2O(1),[Cu4(phen)(OH)4(1,3-BDC)]·(NO32(2),[Cu4(bipy)4(OH)4(H2O)2](1,4-BD C)2·3H2O(3),[Cu4(bipy)4(OH)4(H2O)(1,3-BDC)](1,3-BDC)·13H2O(4),and determined the crystal structure of four copper complexes,copper complex 1 and copper complex 2 The structure of has never been reported before.Since the previously reported copper complexes 3 and 4have similar structures to the complexes in this experiment,in order to explore the relationship between the structure and activity of the complexes,we have also conducted a systematic study on copper complexes 3 and 4.We used ultraviolet-visible spectrophotometry to study the interaction of four copper complexes with CT-DNA respectively,and judge the mode of action between them according to the band changes after they interact with CT-DNA.The experimental results show that:copper The binding constant of complex 1,copper complex 3-4 and CT-DNA is smaller than the insertion binding,so they may interact with CT-DNA in electrostatic binding or groove binding mode.The binding constant of copper complex 2 after interacting with CT-DNA is close to insertion binding,so it may interact with CT-DNA in an insertion mode.We also used the viscosity method to explore the changes in the solution viscosity of the four copper complexes after interacting with CT-DNA,and concluded that the experimental results after the interaction of the two are consistent with the conclusions of the above-mentioned spectroscopic experiments.In addition,we also used gel electrophoresis experiments to explore the nuclease activity of the four copper complexes,and compared the strength of the chemical nuclease activity among them.The results show that copper complexes 1-4 have good The activity of nuclease is as follows:copper complex 2>copper complex 4>copper complex 3>copper complex 1;mechanism experiments show that copper complex 1 breaks plasmid DNA through hydroxyl radicals.
