| Garlic,as a common medicinal and edible plant,is processed into various products,such as black garlic,garlic powder,and garlic slices.A large amount of waste(garlic skin)is generated during the processing and is difficult to use.For this reason,this research uses garlic skin produced during processing as raw material,and extracts the active ingredients from it for use in order to realize the utilization of waste resources.Specifically,the allicin components in garlic skin are determined by high performance liquid chromatography,the extraction process of allicin is optimized,and the extraction amount of allicin is further increased through the microbial pretreatment of garlic skin on the basis of the optimal extraction process.The antibacterial research of plant pathogenic bacteria explores the application potential of garlic peel extract in plant-derived fungicides.The main research results are as follows:First,through single factor experiments,four different extraction methods:organic solvent extraction,ultrasonic-assisted extraction,water extraction,and ultrasonic-assisted water extraction were compared,and it was concluded that organic solvent extraction was the best method.The design of the surface experiment shows that the optimal process conditions for extracting allicin by organic solvent extraction method are 57.28%ethanol solution at 60.37℃ for 7.74 h.The model predicts that under the optimal conditions,the predicted value of allicin is 1.27 × 10-2 mg/g.The actual value is 1.24×10-2 mg/g after experimental verification,which is basically consistent with the predicted value.Secondly,the garlic skin was pretreated with Lactobacillus,Clostridium butyricum,and a combination of both,and the content of allicin became stable after reaching the peak on the 10th,8th,and 10th d,respectively.After the treatment,the amount of allicin extracted in each group was significantly increased,which effectively improved the extraction effect of allicin.Among them,the lactobacillus inoculum had the best effect.Compared with the untreated direct extraction,the amount of allicin extracted was increased by 17%.Finally,the antibacterial effect of garlic peel extracts on Fusarium graminearum and rice sphaerotheca was carried out by the mycelial growth rate method,and the inhibitory rate reached more than 80%.The inhibition of Rhizoctonia graminearum has a good effect.The EC50 values of inhibiting Fusarium graminearum and Fusarium graminearum are 1122.0 mg/L and 1318.3 mg/L,respectively.The garlic peel extract has a more prominent inhibitory effect on Fusarium graminearum.. |
