| Lavender,an angiosperms(Lavandula angustifolia Mill.)plant,rich in various bioactive ingredients such as terpenoids,organic acids and otides,has been widely used in daily chemical products,medicine,family health care,food processing,environmental beautification and other aspects.This paper obtains the best extraction process through the orthogonal test design and response surface analysis,optimizes the purification process of total phenolic acid extract and chemical composition analysis for the development of lavender essential oil waste.The main results are as follows:(1)Lavandula stoechas L,Spike Lavender and white lavender were extracted,the chemical constituents were identified by GC-MS,Through comparative analysis,it was found that linalool content in the extract of lavandula stoechas L was the highest,and Lavandula stoechas L was selected as the follow-up test material.(2)By single factor investigation,orthogonal experimental design and response surface method,the optimal extraction conditions of total phenolic acid of lavender waste were determined as follows: ethanol volume fraction of 60 %,extraction time of 1 h,extraction times of2,solid-liquid ratio of 1:20.By comparing the static adsorption and desorption tests of five kinds of macroporous adsorption resins,the optimal purification process conditions are as follows: Ab-8resin was used,the concentration of loading solution was 0.015 mg/m L,the desorption integral number was 70 %,the p H value of loading solution was 2.0,the p H of desorption solution was 3.0and 2.0 m L/min as the volume flow of loading liquid,and the desorption volume flow was 2.0m L/min.After purification with AB-8 macroporous resin for three times,the total phenolic acid content in lavender waste was about 54.8 %.(3)With Vc as the reference index,the DPPH scavenging rate,reducing power,hydroxyl radical scavenging rate and superoxide anion scavenging rate of total phenolic acid extract of lavender waste before and after purification were compared.It was found that when the sample concentration was 0.7 mg/m L,the reducing power was 0.786 % before purification and 1.125 %after purification.The removal rate of superoxide anion was 12.21 % before purification and32.08 % after purification.The hydroxyl radical scavenging rate was 14.88 % before purification and 31.97 % after purification.When the concentration of the sample was 0.2 mg/m L,the DPPH free radical scavenging rate was 73.21 % before purification and 81.92 % after purification.The antioxidant capacity of total phenolic acid extract of lavender waste after purification was greater than that before purification.The results further verified that the extraction and purification technology was reasonable,reliable and feasible.(4)Identification of lavender waste total phenol acid extract by UPLC-DAD-EIS-MS technology: Benzenediacetic acid,cinnamon acid,rewound polyphenols,tonate,catechphenol. |
